| Objective : It has been reported that Elemene Injection (Elemene Emulsion Injection) can treat lung cancer and other solid tumors, while the mechanisms which treating blood cancer remains unknown. The study was designed to investigate inhibitory effects and mechanisms of Elemene induced apoptosis of K562. We have designed experiments to explore the expression changes and expression mechanism of the cell cyclin inhibitor p27kip1 during apoptosis and analyzed the correlation.Methods : MTT assay was used to examine the changes of the cell roliferation. Microscopic observation and flow cytometry were used to determine the induction of apoptosis and the changes of cell cycle. Immunohistochemistry was performed to determine the protein of p27kip1, and RT-PCR was used to determine the mRNA levels of p27kip1.Results: (1) 12.5～100μg/ml Elemene inhibit proliferation of K562 cells. Elemene and inhibition between time - and dose-dependent manner. (2) In result of TEM , microvillus disappear, electron density of cytoplasm is high, large deciduous apoptosis bodies appear after being treated with 50μg/ml for 48 hours. (3) In result of FCM, after dealt with 50,100μg/ml for 48 hours, cell cycle is controlled in S,and there is a apoptosis peak in DNA tapes.(4) In result of Immunohistochemistry, the protein levels of p27kip1 were gradually upregulated with concentration increasing of Elemene. (5) RT-PCR results show Elemene induced apoptosis in p27Kip1 mRNA levels did not change.Conclusion : The anti-tumor mechanism of Elemene is that it can induce apoptiosis in K562 cells and there is significant correlation with cell cycle inhibitor p27kip1 protein. Elemene may influence the protein expression of p27kip1 in K562 cells, while do not influence their mRNA transcription. p27kip1 protein regulation may happen in post-transcription.
