Objective: To study the effect of RNA interference against HYAL gene on the proliferation and invasion in ovarian epithelial carcinoma cell line SKOV3.Methods: The transcription of HYAL gene was detected by semi-quantitative RT-PCR ; screen to obtain the effective siRNA-HYAL by RT-PCR ;the proliferation was analysed by MTT-assay and flow cytometric; penetrating matrigel capacity of the SKOV3 cells was detected by the pattern of vitro invasion.Result: In SKOV3 cells,the expression of HYAL2 gene is more than HYAL1 gene and HYAL3 gene by semi-quantitative RT-PCR(p<0.05). After RNA interference against HYAL gene, the expression levels of HYAL were decreased significantly. Proliferation was inhibited obviously in the SKOV3 cells by MTT-assay. Flow cytometry showed that cells in S phase were reduced(p<0.05), with more cells were arresting in G0/ G1 phase of cell cycle(p<0.05).And the penetrating matrigel capacity inhibited significantly (p<0.05) . Conclusion: The SKOV3 cells express HYAL2 gene primarily . The capacity of proliferation and invasion can be inhibited by interference against HYAL gene.
