Liver cancer is a familiar malignant tumor in China, winning the third place of malignant tumor incidence, and the second place of malignant tumor death rate. All the traditional clinical therapy is unsuccessful. Chinese medicine can distinctly improve symptoms, and has little side-effect. It is a Chinese characteristic and available path to investigate traditional Chinese medicine, and on the base of which to combine modern medical sciences. Common anti-cancer drugs often destroy normal cells while exerting anti-cancer function. It is a key point to deliver drugs directly to tumor cells, on the condition of not destroying healthy cells, such as liposome and micro-capsule. Nanoparticles are increasingly knockout for their extreme small volume. To use nanoparticles as a carrier for anti-cancer drugs is promising.Zedoary turmeric oil is the effective part of Chinese medicine zedoary, which is prominently effective in anti-cancer and anti-thrombus, and anti-bacteria, protecting liver, etc as well. Anti-cancer is the main pharmacological function. The task prepares ZTO-NP injection with ZTO as modal drug, poly (lactic-hydroxyl acetic acid) (PLGA) as carrier. The purpose is to deliver drug to the target, improve the function and decrease the quantity. It has not been reported both in domestic and abroad.The article has investigated the prescription and preparation of ZTO-NP, quality evaluation, in vivo target of distribution for ZTO and ZTO injection.The key factor of target is the diameter and distribution, exterior quality, embedding ratio(ER) and drug loading (DL).These factors are both important and difficult points in ZTO-NP prescription design and preparation technique. On the bases of single factor test, Uniform Design is adopted to optimize the prescription and preparation. Under the optimum condition, the shape of ZTO-NP is spherical, mean diameter is 233.1nm, zeta potential is-22.1mv, DL is 61.45%, ER is 70.01%, yield is 61.78%. The diameter of ZTO-NP can satisfy the requirement of liver target, and meets the requirement of task design.The task modified and simplified the determination method of Germacrone in biological samples, with which determines the distribution rule and compares the result by injecting ZTO-NP and ZTO through tail vein of mice. With the ratio of the max concentration of drug, the target of ZTO-NP is valued. The result suggests after ZTO is prepared into ZTO-NP, the in vivo distribution is prominently changed, and liver target is achieved.The task firstly investigates the liver targeted drug delivery system from ZTO to ZTO-NP, comparing the in vivo distribution rule. Its subject selection, design, experiment content and method achieve the purpose, and have certain applicable value to liver cancer therapy.
