Research backgroundThe cases of subarachnoid hemorrhage which may occur death after a quarrel or slight injury were frequently encountered in forensic practice. Such cases mostly induced by drunken people. In these cases, the subarachnoid hemorrhage were affected by three factors, such as the diseases, alcohol overtaken and the trauma. It is difficult to assess which is main cause of death often in debate because of the definite mechanism of the mutual impact is uncertain, and a scientific diagnostic criteria can not be provided. In recent years, with application of forensic biomechanics, it could be a approach in forensic research.ObjectiveIn this study, a rabbit model of blood ethanol concentration was established through intragastric administration with gradient ethanol (IGAE). The regional cerebral blood flow and the blood ethanol concentration of each group were measured. The zero-stress state was measured by opening angle with an image processing system. The expression of heat shock protein 70 (HSP 70) in endothelial cells was detected by using immunohistochemical method. In a word, the author hopes to provide a theoretical basis to identify those issues involved in drinking and to provide a new thought and method for solving this kind of issue by using the method of forensic biomechanics. Materials and methods1. Animal grouping and the establishment of the animal model of blood ethanol concentration gradientForty rabbits were randomly divided into four groups. Market selling liquor with 56% alcohol was fed to each animal through intragastric administration with the dosage of 0ml/Kg,5 ml/Kg, 10 ml/Kg and 15 ml/Kg respectively.2. The measurement of the blood ethanol concentration of the animal in each groupThe blood ethanol concentration of the animal in each group was detected by using the gas chromatograph at 30 min, 60 min, 75 min, 90min, 105min and 120min after intragastric administration with liquor.3. The measurement of blood flow in the regional cerebralThe regional cerebral blood flow of each animal in the group was detected in various time after intragastric administration by using the PeriFlux5000 laser Doppler flowmetry. The rabbits were not infused as the control.4. The expression of HSP 70 in endothelial cells of the basilar arteryThe expression of HSP 70 in endothelial cells of the basilar artery was observed with the immunohistochemical staining, and an image processing system was used to quantitate the relative expression of HSP 70 in each group.5. Detection of the zero-stress state of the basilar arteryThe basilar artery was taken out 2 hours after intragastric administration with liquor, the artery was cut into rings about 1mm in length, then cut the rings longitudinally to free the tension, and put them in the kreb's fluid for 20 minutes. The geometrical information of artery rings was collected by using the digital camera. The zero-stress state characterized by opening angle was analyzed with an image processing system named AutoCAD Electrical 2006.Result 1. The blood ethanol concentration of each group increased after IGAE, and came to the peak at 1.5-2h after IGAE. The value of blood ethanol concentration has a positive correlation with the dosage of intragastric administrated liquor.2. The regional cerebral blood flow of each group increased after IGAE. The value of the regional cerebral blood flow came to the peak at 30 min after IGAE, and was stable from 30min to 60min. The regional cerebral blood flow has a positive correlation with the dosage of intragastric administrated liquor.3. The expression of HSP 70 increased as the increase of the dosage of intragastric administrated liquor. There was statistically difference of the optical density(IOD) of the stained slides in each group (P <0.05).4. The opening angel decreased as the increase of the liqor from IGAE. There was statistically difference of the opening angel in each group (P <0.05). The degree of the opening angel has a negative correlation with the dosage of intragastric administrated liquor.ConclusionIt is suggested thatâ‘ The value of blood ethanol concentration has a positive correlation with the dosage of intragastric administrated liquor.â‘¡The regional cerebral blood flow has a positive correlation with the dosage of intragastric administrated liquor.â‘¢The expression of HSP 70 increased as the increase of the dosage of intragastric administrated liquor.â‘£The degree of the opening angel has a negative correlation with the dosage of intragastric administrated liquor.
