Objective To investigate the significance of expression ofperoxisome proliferator-activated receptor_Y(PPAR_Y) andcyclooxygenase-2(COX-2) in gastric carcinoma and to identifythe correlation between PPAR_Y and COX-2.Methods Strept-Avidin Biotin Complex immunohistochemicalmethods were adopted to examine the expression of PPAR_Yand COX-2 in 40 patients with gastric adenocarcinoma,compare with their paired adjacent mucosa and normalmucosa.Results The positive rate of PPAR_Y protein and COX-2protein both were significantly higher in gastriccarcinomas(70%,72.5%) than that in their paired adjacentmucosa(25%,35%) and normal mucosa(17.5%,5%). Fromnormal gastric mucosa, adjacent gastric mucosa to gastriccarcinoma, expressions of PPAR_Y and COX-2 showed anascending tendency (P<0.01, P<0.01).In gastriccarcinoma, expressions of PPAR_Y only were relative withtumor cell differentiation (R=0.769,P<0.01). However,expressions of COX-2 were relative with depth of invasion,lymph node metastasis and clinical stage (R=0.828,P<0. 01).There was a significant correlation between expressions ofPPAR_Y and COX-2 in gastric carcinoma (r_s=0.560, P<0.01).Conclusion PPAR_Y and COX-2 protein overexpression is arelatively early event in gastric carcinogenesis, and it may playboth an independent and synergetic role in progression ofgastric carcionoma.
