Objectives: 1. To detect the protein expression levels of TRAIL, DR5and cFLIP in human RCC and normal renal tissue; 2. To compare theexpression difference of TRAIL, DR5 and cFLIP between RCC and normalrenal tissue. To compare the expression difference of TRAIL, DR5 and cFLIPin RCC with different clinical and pathologic characters. To investigate theclinical significances of the expression difference.Methods: The protein expression levels of TRAIL, DR5 and cFLIP wereevaluated by immunohistochemistry (S-P method) in the tissues of 50 humanRCC and 40 normal renal tissues. The expression difference of them in RCCand normal renal tissue was respectively analyzed. The expression differenceof them in RCC with different clinical stage and patho-grade was respectivelyanalyzed.Results: 1. TRAIL and DR5 were mainly stained in cell membrane whilecFLIP were mostly stained in cytoplasms. 2. Twenty-two cases showedpositive staining of TRAIL with 44% of positive rate and 0.9±0.4 ofimmunoreactive score(IRS) in 50 RCC; Twenty-one cases showed positive staining of TRAIL with 52.5% of positive rate and 1.2±0.6 ofimmunoreactive score(IRS) in 40 normal renal tissues. Thirty-four casesshowed positive staining of DR5 with 68% of positive rate and 1.4±0.7 ofimmunoreactive score(IRS) in 50 RCC; Fiffeen cases showed positivestaining of DR5 with 37.5% of positive rate and 0.7±0.2 of immunoreactivescore(IRS) in 40 normal renal tissues. Twenty-five cases showed positivestaining of cFLIP with 50% of positive rate and 1.5±0.7 of immunoreactivescore(IRS) in 50 RCC; Fourteen cases showed positive staining of cFLIP with35% of positive rate and 1.2±0.8 of immunoreactive score(IRS) in 40normal renal tissues. 3. The expression difference of DR5 between RCC andnormal renal tissue was significant in statistics (p=0.0060).The difference ofTRAIL between RCC and normal renal tissue was not significant (p=0.5250)and so did cFLIP(p=0.2000). 4. The expression difference of DR5 in RCCwith different patho-gades was significant in statistics (p=0.031). Theexpression difference of DR5 in RCC with different clinical stage anddifferent tumor volume was not significant (p=0.903; p=0.631)。Conclusions: 1. A high expression level of DR5 was detected in RCCand a low expression level of DR5 was detected in normal renal tissues. Theexpression difference of TRAIL and cFLIP between RCC and normal renaltissues were not significant; 2. The expression difference of DR5 in RCCwith different patho-grades suggested The expression level of DR5 wasprobably related with the progression of RCC; 3. The expression differenceof TRAIL and DR5 between RCC and normal renal tissue indicated that themechanism of apoptosis induced by TRAIL may not play an important role inmolecular level in the morbidity of RCC.
