Objective To discuss the effect of c-Flip gene expression in TRAIL-induced apoptosis in breast cancer cell line MCF-7.Methods1.Chosing breast cancer cells MCF-7 to cultivate and passage.2.Transfect MCF-7 cells with recombinant adenoviruses Ad-cFLIP siRNA/Ad-sTRAIL,alone or combined.3.Real-time quantitative RT-PCR was used to detect the expression level changes of cFLIP mRNA and TRAIL mRNA in MCF-7 cell after transfection.4.MCF-7 Cell cytotoxicity activity was detected by MTT assay and Crystal violet staining method.The apoptnsis of MCF-7 cells was detected in Hoechst33258 kits.Results1.Compared with the negative control group,the c-FLIP relative expression of the c-FLIP-siRNA group and the c-FLIP-siRNA+TRAIL group respectively was(0.34 ± 0.16)and(0.39 ± 0.48)times;the TRAIL relative expression of the TRAIL group and the c-FLIP-siRNA + TRAIL group respectively was(96.21 ± 1.54)and(87.33± 1.66)times.2.The inhibition rate(%)of TRAIL group?c-FLIP-siRNA group and TRAIL+c-FLIP-siRNA group respectively(60.27± 1.25),(11.34± 1.74)and(74.91 ±2.12).3.Compared with the negative control group(3.12 ± 1.54),the apoptosis rate of TRAIL group(8.79 ± 2.46)?c-FLIP-siRNA group(4.85 ± 2.82)and c-FLIP-siRNA+TRAIL group(17.50± 3.17)Increased significantly.Conclusionc-FLIP gene suppression with siRNA interference can obviously promote TRAIL-induced breast cancer cell apoptosis in MCF-7 cell line.
