Objective: To establish the model of yin deficiency hyperthyroidism by fed rat with thyroid. Observe the effect of Yinning tablets on the treatment of in yin deficiency hyperthyroidism in rats. Detect and analysis the serum tri-iodothyronine (T3), thyroxine (T4), free tri-iodothyronine (FT3), free thyroxine (FT4), thyroid-stimulating hormone (TSH) of yin deficiency hyperthyroidism rats', Record rats' appearance, weight, temperature, heart rate, patho-histological, and invest the toxicological effect of Yinning tablets. Establish an objective, accurate and reliable foundation for clinical trials.Methods:(1) model of yin deficiency hyperthyroidismThirty-five male Wistar rats were randomly divided into seven groups (n=5), the first one was control group, the 2 to 7 group was the model groups. Each model rat was firstly ig thyroid powder2.5mg/kg.d. The control group was ig with same volume NS. Groups 2~6 were executed after ig drug 20 days 30 days, 40 days, 50 days, 60 days.The first group and the 7 group were executed after 70 days. Observe the model rats appearance, behavior, weight, temperature, heart rate, Test serum T3, T4, ET4, ET4 and TSH levels.(2) the effect of Yinning tablets on the thyroid function, the general condition and the pathological change of yin deficiency hyperthyroidism96 Wistar rats were randomly divided into six groups (N=16): normal control group; model control group; Jia Kangling group; low-dose, mid-dose and high-dose Yinning group. After the model was successfully established, Jia Kangling (0.7g/kg), Yinning films (0.225g/kg. 0.45g/kg, 0.9g/kg, each was 5, 10, 20 times as much as clinical dosage) were given one time a day. Model and normal control groups were orally given NS. Eight rats of each group were randomly executed one month, two months later after treatment. Observe rats' appearance, behavior, weight, temperature, heart rate, test serum T3, T4, FT3, FT4 and TSH. Then thyroid tissues of all animals were gotten, which were preserved in formalin solution for pathological test.(3) Experimental ToxicologyAfter one night fasting, 20 Kunming mice were given (two times a day) the largest concentration of the tested drug preparation (0.5g/ml) by ig, each dose was 22.25g/kg, the total dosage was 44.5g/kg/d at 12 hour' interval. Observe the toxic symptoms and record the dead number of mice after orally provided Yinning for seven consecutive days.Results: The model of yin deficiency hyperthyroidism by fed rat with thyroid is successful, Yinning tablets and Jia Kangling play important treatment role in hyperthyroidism. After fed thyroid for 40 days, the serum T3, T4, FT3, FT4 of experimental rats were significantly higher than the normal control group (P<0.001); while serum TSH significantly lower than that (P<0.001), at the end of experiment, Most of experimental rats displayed high metabolism symptoms of yin deficiency hyperthyroidism such as tremor, emaciation, dim hair, weight loss, accelerated heart rate, fervescence and so on. When treated with Yinning and Jia Kangling, the serum T3, T4, FT3, FT4 and rectal temperature of model rats' were significantly decreased(P<0.01). Serum TSH values were significantly rised (P<0.01), heart rates were decreased. Of all yin deficiency hyperthyroidism rats treated with Yinning for two months, from the rat body weight, heart rate, body temperature to serum T3, T4, FT3, FT4, TSH, mid-dose Yinning group and the normal group were mostly similar. As the maximum tolerated dose experiments, all the mice survived for seven days, activity and mobility, spirit and hair color were normal, drink and eat well without tremor and nose, eyes, mouth abnormal secretion. No abnormality was found in intemal organs by autopsy.Conclusion: To establish the model of yin deficiency hyperthyroidism by fed rat with thyroid is success. Yinning tablets have significant therapeutic effect on the rats model with yi-n deficiency hyperthyroidism. Yinning tablets are safe in the dose of treatment. The therapeutic effects of different doses and course of treatment of the Yinning tablets on experimental rats with yin deficiency hyperthyroidism were Preliminarially studied. It provide an experimental foundation for the clinical treatment, research and development of new drugs on the hyperthyroidism.
