Screening And Isolation Of α-glucosidase Inhibitor From Microbial Metabolites And The Inhibitor-producing Strain's Identification

Diabetes is a kind of disease reason for metabolic disorders,the character of it is postprandial hyperglycemia.Alpha-glucosidase inhibitor represents a new concept for the treatment of diabetes.It slows the absorption kinetics of dietary carbohydrates by inhibition of alpha-glucosidase activity,and so reduces the post-prandial hyperglycemia increment and treat other dumping syndrome.This paper try to screening new Alpha-glucosidase inhibitor from the natural microbe metabolites, that may be used as medicine or the leading compound for treatment diabetes.The content and creative fruit of this paper mainly included four parts as followed.1.Based on other scientist's study,we had built an effective model for screening Alpha-glucosidase inhibitor,by this model,we got 10 microorganism strains from 3000 strains which fermentation broths can strongly inhibit the active of Alpha-glucosidase.2.One of them,strains number is 506157,with the strongest inhibitory activity,was selected to research deeply.The active substance was isolated from its fermentation broths by HD-1 ion-exchange-resin-column,HZ806 adsorption resin-column,TLC, HPLC and preparative HPLC,this compound was obtained as a white powder.3. This compound demonstrated a potent inhibitory effect on alpha-glucosidase activity,the activity of 506157 is about ten times stronger than acarobose.We identified its structure by LC/MS,US,1HNMR,13CNMR,its molecular quality is 254,it is an isoflavone, the chemical name is 4',7-dihydroisoflavone(daizein).4.We identified this strains by its shape character,cultivate character,physiological and biochemistry character,and by using Homology Analysis of 16SrDNA sequencing, we confirmed its Streptomyces,it is similar to Streptomyces albulus.
