Objective:The hepatocellular carcinoma (HCC) is one of the most common malignant tumors in our country, and neoangiogenesis are closely involved in the invasion and metastasis of HCC.So to identify the principium of invasion and metastasis and to search effective approaches to prevent the recurrence and metastasis of HCC remain a challenge to modern medical science.By observing the expressions of HIF-1αand VEGF in seeding carcinoma.To discuss the relationship of VEGF and HIF-1αexpression with the feature of invasion and metastasis in HCC tissues and between VEGF and HIF-1α.Also observe the inhibition of tumor Angiogenesis by octreotide and cooperate with the HAL,consummate its antineoplastic mechanism,basis for clinical application.Methods:As to create liver cancer model, Walker-256 carcinoma cells were transplanted into rat's liver. Hepatic arterial ligation ( HAL) was used to block the hepatic arterial blood supply and simulated TACE.A-HAL plus octreotide group,B-HAL group,C-octreotide group,D-control group. Tumor tissues were excised after therapy was evaluated. The expression of VEGF and HIF-1αwas detected by immunohistochemical in tumor tissue.Results:1. The metastasis of hepatic tumor(pulmonary metastasis,the liver recurrent,ascites).In group D were decreased significantly compared with group B(P<0.05), and that in group A were also decreased significantly compared with group B(P<0.05). 2. Expression of VEGF in every group : in group A was downregulated than that in group B(P<0.05);and that in group C also downregulated than that in group D(P<0.05);In group D decreased significantly compared with group B(P<0.05);and however,there is not difference significance in group C and A(P>0.05).3. Expression of HIF-1αin every group:in group A was smaller than that in group B(P<0.05);and that in group C smaller than that in group D,but the difference had no significance(P>0.05);In group D decreased significantly compared with group B(P<0.05);in group A smaller than that in group C, but the difference had no significance(P>0.05).4. There was a positive correlation between HIF-1αand VEGF in Seeding carcinoma (R=0.574,P=0.008), more increased than after treated with HAL.(R=0.696,P=0.001).Conclusion : 1. HAL aggravates hypoxia in tumor, and promotes the expression of HIF-1αand VEGF protein, which could stimulates the metastasis of hepatic tumor.2. The expression of VEGF were upregulated by HIF-1,specially in hypoxia.3. Octreotide could downregulated the expression of VEGF and HIF-1αin tumor tissue, which reduced the metastasis of tumors.
