Objective:To investigate the expression of hypoxia inducible factor-1a(HIF-1a)and vascular endothelial growth factor(VEGF)and its receptor-1(sFlt-1)in placenta in preeclampsia.Methods:72 cases of preeclampsia were collected from June 2016 to May 2017.34 cases of severe preeclampsia(severe preeclampsia group,group c),38 cases of mild preeclampsia(mild preeclampsia group,group b).35 normal late pregnant women who delivered in obstetrics department of our hospital during the same period were collected as control group(group a).The expression of HIF-1 alpha in placenta of three groups was detected by immunohistochemical SABC and Western blot(W-B),The levels of serum vascular endothelial growth factor and sFlt-1 were detected by ELISA,and urine vascular endothelial growth factor and sFlt-1 were also detected.Results:1.Immunohistochemical results showed that the expression of HIF-1 alpha in placenta of group C and group B was higher than that of group a(P < 0.01),and the expression of HIF-1 alpha in group C was significantly higher than that in group B(P < 0.05).2.The results of W-B method showed that the expression of HIF-1 alpha in placenta tissue of group B and group C increased significantly compared with group a(P < 0.01,P < 0.01);Compared with group b,the expression of HIF-1 alpha in placenta tissue of group C increased significantly(P < 0.01).3.Compared with group a,the serum levels of vascular endothelial growth factor in group B and C decreased significantly(P < 0.01,P < 0.01),and sFlt-1 increased significantly(P < 0.01,P < 0.01);Compared with group b,the serum level of vascular endothelial growth factor in group C was significantly lower(P=0.009)and sFlt-1 was significantly higher(P<0.01).Person correlation analysis showed that the expression of HIF-1a in placenta of preeclampsia was negatively correlated with serum level of vascular endothelial growth factor,and positively correlated with sFlt-1(P < 0.05).4.Compared with group a,the levels of urinary vascular endothelial growth factor(VEGF)in group B and group C were significantly lower and sFlt-1 was significantly higher(P < 0.01);compared with group b,the levels of urinary vascular endothelial growth factor(VEGF)in group C were significantly lower and sFlt-1 was significantly higher(P < 0.01).5.Serum level of vascular endothelial growth factor was positively correlated with urine level of vascular endothelial growth factor,negatively correlated with urine level of sFlt-1(P < 0.01);serum level of vascular endothelial growth factor was negatively correlated with urine level of vascular endothelial growth factor,and positively correlated with urine level of sFlt-1(P < 0.01).Conclusion:1.HIF-1 alpha plays a role in the occurrence and progression of preeclampsia.2.Serum vascular endothelial growth factor and sFlt-1 are involved in the occurrence and progress of preeclampsia,and they may be biological indicators for predicting the occurrence and development of preeclampsia.3.Detection of sFlt-1 in urine of pregnant women is expected to be a marker for clinical diagnosis of preeclampsia.
