| Objective The aim of this study was to investigate the MDR-reversing effect ofChinese medicine Extractum trametes robiniophila murr on human hepatocellularcarcinoma HepG2/ADM in vitro and its potential mechanism. Methods MTTassay was used to test the toxicity of the Chinese medicine Extractum trametesrobiniophila murr and the chemosensitivity to chemotherapeutics in Chinese medicineExtractum trametes robiniophila murr-treated HepG2 and HepG2/ADM cell lines.Flow cytometry was used to determine intracellular ADM (Adriamycin) concentration.The expression of MDR1 was measured by RT-PCR. Results The inhibition rates ofHepG2 and HepG2/ADM caused by Chinese medicine Extractum trametes robiniophilamurr were less than 10% under the dose of 0.01g/L, and the IC50 were 0.917g/L and1.66g/L. Chinese medicine Extractum trametes robiniophila murr(0.008g/L) partlyovercame the MDR of cell line HepG2/ADM and decreased the drug resistance. TheIC50 of ADM, cisplatin (DDP), mytomycin (MMC), 5-fluororacil (5-FU) was descendedobviously while with Chinese medicine Extractum trametes robiniophilamurr(0.008g/L). The relative reverse efficiencies to ADM, DDP, MMC, 5-FU were 79%, 73.5%, 97.4% and 86.7%, respectively. While, after exposure to 0.008g/LChinese medicine Extractum trametes robiniophila murr 4 hours, the concentration ofADM in HepG2/ADM was significantly increased(P＜0.05). And exposure to 0.008g/LChinese medicine Extractum trametes robiniophila murr 48 hours, the MDR1expression in HepG2/ADM cells decreased 33.7% by the RT-RCR assay, still higher than in HepG2 cells. Conclusion Chinese medicine Extractum trametesrobiniophila murr can reverse the multi-drug resistance of human hepatocellularcarcinoma in vitro. The possible mechanism is related to down-regulating theexpression of MDR1 and raising the concentration of drug inside the cells. It canincreases the sensitivity of chemotherapy for cancer patients, and provide the theorybasis for combined chemotherapy.
