| Objective To prepare PLGA microspheres containing recombinant human growth hormone and study these sustained release in vitro and in vivo. Methods HSA microspheres were prepared by a double-emulsification solvent evaporation method. To design orthogonal experiment to optimize the preparation condition of rhGH-loaded microspheres and study characters of optimal rhGH-loaded microspheres. Results The diameters of optimal rhGH-loaded microspheres were mainly between 10μm and 40μm. The encapsulation efficiency and drug loading were (85.25±2.5)% and (11.52±0.42)% respectively.It showed good lente liberantes effect in vitro and in vivo. Conclusion This technique is simple and has a good reproducibility. It is a fine sustained dosage form for recombinant human growth hormone. |
