Objective To study the expression of visfatin mRNA in abdominal subcutaneous and omental adipose tissue of obesity and type 2 diabetes mellitus to evaluate its correlation with insulin resistence (HOMA-IR) and other agents.Methods Total mRNA was extracted from visceral and abdominal subcutaneous adipose tissue. Semi-quantitative RT-PCR was employed to study the expression of visfatin mRNA in adipose tissue of 20 cases with obesity, 22 cases with type 2 diabetes and 20 controls. Meanwhile the anthropometric data such as weight, waist circumferencem, hip circumferencem were measured. Fasting plasma levels of insulin (FINS), fasting plasma glucose, lipids were determined . HOMA-IR was calculated.Results 1. BMI, LDL, SBP, WHR were significantly higher in obesity group than those in controls(P<0.05 or P<0.01). WHR, FPG, HOMA-IR were significantly higher in diabetes than those in controls (P<0.05 or P<0.01). 2. In controls, diabetes and obesity group, the visfatin mRNA expression in omental adipose tissue were significantly higher than those in subcutaneous adipose tissue respectively (P<0.01). The visfatin mRNA expression in omental adipose tissue in diabetes and obesity group was significantly higher than in controls (P<0.01). In controls, diabetes and obesity group, the visfatin mRNA expression in subcutaneous adipose tissue were not significantly different (P>0.05). 3. Pearson correlation analysis showed that the expression of visfatin mRNA in VAT was positively correlated with HOMA-IR and the expression of visfatin mRNA in SAT (P<0.05), that the expression of visfatin mRNA in SAT was positively correlated with the expression of visfatin mRNA in VAT (P<0.05). 4. Multi-stepwise regression analysis demonstrated that the expression of visfatin mRNA in SAT was significantly correlated with the expression of visfatin mRNA in VAT, that the expression of visfatin mRNA in VAT was significantly correlated with HOMA-IR and the expression of visfatin mRNA in SAT.Conclusion 1. The expression of visfatin mRNA in VAT is higher than it in SAT. 2. The expression of visfatin mRNA in VAT in diabetes and obesity group is higher than in controls (P<0.01). 3. The expression of visfatin mRNA in VAT is correlated with HOMA-IR. The expression of visfatin mRNA in VAT seems to be an important parameter of insulin resistance. |
