Objective: This experiment mainly observed impacts of the Ku Suan TongTiao Fang on Spontaneously type 2 diabetes mellitus in rats with insulin resistance on glucose and lipid metabolism,and Fatty acid synthase(FAS) protein expression in abdominal fat.Methods: In 30 male Zucker Diabetes Fat(ZDF) rats as experimental subjects, the use of high-fat diet fed the rats until twelve weeks to form type 2 diabetic rats.They were randomly divided into three groups, namely, model group, Pioglitazone treatment group, KuSuan TongTiao Fang treatment group.For the control group of non-diabetic male with 10 Zucker Lean(ZL) rats, they were fed normal diet.Weight changes were recorded during the experiment,and after 12 weeks of continuous drug treatment, blood glucose, serum insulin(INS), lipids four terms(TC,TG,LDL-C,HDL-C),Hb A1 C,etc were determined; and by western blot method,the abdominal adipose tissue of rats FAS expression of protein were detected.Results: 1.Compared with the normal group,FBG,HbA1 C and INS of model group were significantly increased(P<0.05).Compared with the model group,INS of pioglitazone group significantly reduced(P<0.05);KuSuan TongTiao Fang group revealing FBG, HbA1 C, INS were significantly lowered,than the model group,which INS close to pioglitazone group.2.Compared with the normal group,levels of serum TG,TC,LDL-C,HDL-C of model group were significantly different(P<0.01). Compared with the model group, TG, TC of pioglitazone group significantly reduced(P<0.01); levels of TG, TC, HDL-C, LDL-C of KuSuan TongTiao Fang group compared with the model group,the difference was statistically significant.3.After 12 weeks of drug treatment,body weight of the model group, pioglitazone group and KuSuan TongTiao Fang group were significantly higher than the control group(P<0.01).8 weeks or 12 weeks after Ku Suan TongTiao Fang group significantly decreased body weight compared with the model group.There was no significant difference between pioglitazone group and the model group(P>0.05).4.The expression of FAS protein in abdominal adipose tissue:Compared with the normal group,FAS protein expression of model group was significantly increased(P<0.05).Pioglitazone group and KuSuan TongTiao Fang group significantly reduced FAS protein expression, compared with model group had significantly improved(P<0.01).Conclusion:1.KuSuan TongTiao Fang has hypoglycemic,lipid-lowering,loseweight,improve insulin sensitivity and improving insulin resistance.2.KuSuan TongTiao Fang group could significantly reduce the FAS protein expression of abdominal adipose tissue, the experimental results show that lipid disorders with type 2 diabetes are associated with FAS.KuSuan TongTiao Fang inhibites FAS by a key enzyme of the parties synthesized body fat pathway,reduces fat deposition, controls fat synthesis, loses weight. FAS may be the role of KuSuan TongTiao Fang through a new target side,and the mechanism may interfere with the synthesis and catabolism of body fat tissue, thereby improving insulin resistance. |
