Background: Infrasound is a kind of mechanical vibration wave with frequency between 0.0001~20Hz. And its characteristic included low frequency, long Wave Length, slow fading, far spreading and strong penetrating power etc.Infrasound could extensive act each organ and tissue through biologic resonance. So it could generate widely biological effect, included organs, tissues, cells and molecule. The influence of infrasound correlated with pressure, frequency and action time of it. In the mechanism of action, the primary injury of cell structure and function was the most important point. Previous studies indicated that the injury of organism had positive rate with infrasound pressure level. Now, most studies about infrasound concentrated on the influence on normal tissues and cells.Recent years some researchers found that low frequency infrasound could make the activity of antioxidation enzyme increased. So it had huge potential at the aspect of desease diagnosis and therapy. The infrasound 8~TM made by Chi corporation (USA) could generate low frequency infrasound (4~20 Hz, 79.75~86.11 dB). The function of Infrasound had two aspect, one is the injury of normal tissues, the other is to be a clinical treatment.At present, there were no internal experiments and studies about ex vivo cells which had been acted by infrasound, otherwise it was the first study which used the infrasound 8TM treatment implement to act the carcinoma cells and observed the influence on cells' biological feature.Objective: Studied the influence of HL-60 cells acted by the infrasound 8~TM generator, and provided the experiment evidence for clinic therapy.Methods: Use the infrasound emitted by infrasound 8~TM generator (NO.3 mode) to directly act HL-60 cells (as effect group) and respectively detecting after 15min, 30min, 60 min, 90min and 120min, normal HL-60 cells (cultured into 10%NBCS RPMI-1640 medium) were act as control group. Trypan blue assay was used to detecting cytoactive, MTT assay was used to detecting cell proliferation, flow cytometry was used to detecting apoptosis, and scanning electron microscope was used to observe cell morphous.Results:Cell proliferationTrypan blue assay: The result displayed that the cells of control group and effect group (after act 15 min, 30 min, 60 min, 90 min and 120 min) were persistent growing at culture Oh, 24h and 48h, and effect group had no difference with control group (P>0.05). Each group had few chromophobe, cell survival rate were all upon 95%.MTT assay: there had no difference between effect group and control group (P>0.05). It was much clear that the cell number of 120min effect group was lower than control group's, but the other action times were not.ApoptosisAnnexin V-EGFP / PI: The two groups' cells were acted 120min by infrasound and respectively detecting after cultured 24hrs and 48hrs. Living cells in each group was about 90%, the total apoptosis were all 10%, the act group and control group had no difference.Cell morphology Fluorescent staining: The number of bright blue fluorescent light in each group had no difference.Scanning electron: Control group: cell surface was not flat, the cell bloom was very dense. Effect group: cell surface was very flat, the cell bloom was very sparseness.Conclusion: The cells acted by infrasound hadn't primary injury. And the cell proliferation and apoptosis hadn't any influence. The mode of action on carcinoma cells of infrasound was mechanical vibration, it could effect morphous of cell membrane, for instance, it could make cell membrane elevatio more shallow than normal cells', could make microvilli and ciliary shed. Then the cell internal environment was changed. The infrasound from the infrasound 8TM generator (Chi) could obviously affect cell morphous, but it couldn't induced HL-60 cells' primary injury.
