ObjectiveThis study was to investigate the effect of conditioned medium from two types of human breast cancer lines on cell morphous, proliferation and migration of MG-63 cell, the effect of conditioned medium from MG-63 cell on morphous, proliferation and migration of two types of human breast cancer lines.MethodsMG-63 cell were treated with conditioned medium from MDA-MB-231 or T47D breast cancer cells. Proliferation was measured by CCK-8 assay. Breast cancer cells(MDA-MB-231 and T47D) were treated with conditioned medium from MG-63 cell.Proliferation was measured by CCK-8 assay. The effect of conditioned medium from MDA-MB-231 or T47D breast cancer cells on MG-63 cell spreading was detected by cell sctatch test. The effect of conditioned medium from MG-63 cell on MDA-MB-231 or T47D breast cancer cells spreading was detected by cell sctatch test.Results1. The inhibition effect of conditioned medium from MDA-MB-231 or T47D breast cancer cells on osteoblasts.(1) Cell morphous:When MG-63 cell were treated with conditioned medium from MDA-MB-231 or T47D cells, the cells became long and spindle-like,assumed a fibroblastic morphology.(2) Proliferation:Conditioned medium from MDA-MB-231 or T47D breast cancer cells has proliferation inhibition effect on MG-63 cell. The highest proliferation inhibition rates were 50%. From 1 to 6 day, the OD of MG-63 cell treated with conditioned medium from MDA-MB-231 or T47D cells have statistically significant difference(P<0.05).Moreover, comparison of the proliferation inhibition rates of culture with 100% MDA-MB-231 and T47D cell supernatants on MG-63 cell from 1d to 6d, the proliferation inhibition rates of group A were higher than group B.(3) Migration:Conditioned medium from breast cancer cells inhibited migration of MG-63 cell,compared with those cultured with vehicle control medium. 2. The highest proliferation inhibition rates were 40%. From 1 to 6 day, the OD of MDA-MB-231 and T47D cells treated with conditioned medium from MG-63 cell have statistically significant difference (P<0.05). Moreover, comparison of the proliferation inhibition rates of culture with 100% MG-63 cell supernatant on MDA-MB-231 and T47D cells from 1d to 6d, the proliferation inhibition rates of group a were higher than group b.(3) Migration:Conditioned medium from MG-63 cell inhibited migration of breast cancer cells,compared with those cultured with vehicle control medium.Conclusions1. This results suggest that human breast cancer cells inhibit proliferation, adhesion and spreading acitivity of MG-63 cell. The proliferation inhibition effects of conditioned medium from MDA-MB-231 cell were stronger than T47D cell.2. MG-63 cell inhibit proliferation, adhesion,and spreading acitivity of human breast cancer cells. The proliferation inhibition effects of conditioned medium from MG-63 cell on MDA-MB-231 cell were stronger than on T47D cell.
