Objective: The purpose of this study was to find the markers which can be used in early adjuvant diagnosis of gastric adenocarcinoma (GAC).Methods: 117 operative tissues were analyzed using fluorescence in situ hybridization (FISH) and immunohistochemistry (IHC), including 99 GAC and 18 adjacent normal mucosae. For FISH analysis, we selected two centromere probes cen8 and cen17 specific for chromosome 8 and 17, which frequently amplified in GAC according to the references. For IHC staining, TP53 and TOP2A were detected using tissue microarray (TMA) constructed with above samples. Data were analyzed by SPSS13.0 statistical software for Windows.Results: We observed that 65.7% ofcen8 and 58.6% of cen17 were aneuploid in tumor tissues. 45.5% of TP53 and 83.8 % of TOP2A were overexpressed in GAG samples (p<0.000 compared with normal mucosa). There existed trisome simultaneously in three out of 18 tumors and adjacent mucosae. Aneuploid of cen8 and cen17 were more frequent in grade 1 or 2 of GAC than grade 3 (p<0.05). A significant correlation between aneuploid of cen17 and cardia was found (p<0.05). There is no correlation between aneuploid of ten8, cen17 and age, sex of patients, lymph node metastasis, and clinical stage of tumors. Overexpression of TP53 protein was associated with stage and size of tumors (p<0.05). Additionally, a negative correlation was observed between overexpression of TOP2A and lympho node metastasis (LNM) as well as TNM classification (p<0.05).Conclusions: Detection of aneuploid of cen8 and cen17, overexpression of TP53 and TOP2A may help the diagnosis of GAC. Aaneuploid of cen8 in adjacent mucosa may have a predictive role in tumor recurrence and might be used as a candidate marker for the early diagnosis of gastric adenocarcinoma.
