FGF2,IGF-1R And Ki-67 Expression In Olfactory Epithelium Of Patients Suffering From Dysosmia Caused By Chronic Sinusitis

Objective: To study the pathogenesis of dysosphresia in patients with chronic sinusitis,to discuses the expression and significance of FGF2,IGF-1R and Ki-67 in olfactory mucosa .Methods: Forty-three patients ranging in age from 20 to 54 years underwent FESS in the period from September 2005 to February 2006, and average age was 34 years. Twenty-three cases were male, while eleven cases were female. They had no histories of trauma in head and neural diseases. Before operating, their olfactory functions were examined with CCCRC. According to their CCCRC scores, they were distributed two groups. Group A: Patients with chronic sinusitis and dysosmia (CCCRC score<6.0, no obvious deviation of nasal septum) 32 cases; Group B: Patients with deviation of nasal septum and a normal olfactory function (CCCRC score≥6.0, Computered Tomography (CT) scan without evidence of chronic sinusitis) 11 cases. With the agreeing of all the patients, olfactory mucosas of the nasal septum vs superior turbinate were obtained at the time of surgery. And FGF2,IGF - 1R and Ki-67 were examined by the method of immunohistochemistry.Tissue sections were observed under optical microscope.Results:Histopathology Group A obviously became thinnest, the number of ORNs distinctly reduced, and a severe inflammation infiltrated in the lamina propria; FGF2,IGF-1R which were stained in kytoplasm on olfactory mucosa of group A, was evidently lighter compared to group B (p<0.05); Ki-67 which were stained in nucelus on olfactory mucosa of group A, was evidently lighter compared to group B (p<0.05). There was positive correlation between positive cells of Ki-67 and Stain density of FGF2 of group A (r=0.380 p<0.05); There was positive correlation between positive cells of Ki-67 and Stain density of IGF-1R of group A (r=0.436 p<0.05);And There was positive correlation between Stain density FGF2 and Stain density of IGF-1R of group A (r=0.777 p<0.01).Conclusion: Olfactory mucosas of patients with chronic sinusitis and dysosmia was destroied obvisouly . FGF2 and IGF - 1R that promoting of regeneration in the olfactory epithelium decrease notably of group A. Ki-67 also decrease obviously .These elucidated that olfactory mucosas of patients with chronic sinusitis and dysosmia were damaged by inflammatory infiltration ,so can not produce enough ORNs ,due to dysosmia.
