Objective:Rat glomerular mesangial cells(MC) induced by oxidized low-density lipoprotein(OX-LDL)were treated with Panax notoginseng saponins(PNS) of different concentrations. To detect the expressions of transforming growth factor– beta1( TGF -β1) and monocyte chemo-attractant protein-1(MCP–1),observe the translocation of nuclear transcriptional factor of KappaB(NF-κB),and to explore the possible protective effect and mechanism of PNS in lipid-induced glomerular injury.Methods:A rat glomerular mesangial cell line cultured in vitro was induced by OX-LDL of different doses. MC exposed in OX-LDL was treated with PNS of different concentrations. Semi-quantitative reverse transcription polymerase chain reaction(RT-PCR) was used to detect the expressions of TGF-β1 and MCP-1 mRNA. Enzyme linked immunosorbent assay(ELISA) was used to determine the levels of TGF-β1 and MCP-1 protein in the supernatant. And the translocation of NF-κB p65 was observed by immunocytochemistry.Results:OX-LDL significantly increased the TGF-β1 and MCP-1 expressions of MC in dose-related manner. After adding different concentration PNS with certain action time,the expression levels of TGF-β1 and MCP-1 in MC remarkably decreased compared with those in cells induced by OX-LDL. NF-κB activity markedly increased after stimulation by OX-LDL,the translocation rate of p65 from cytoplasm to nucleus was higher than that in the control group.This effect could be inhibited by PNS.Conclusions:OX-LDL could promote the expressions of TGF-β1 and MCP-1 in MC. PNS could inhibit this promotion of OX-LDL. This inhibition may affect on transcriptional level. The influence of PNS on the activation of NF-κB and translocation of p65 may be one of the mechanisms.
