Expression of MCP-1 in human mesangial cells induced by Ox-LDL is mediated by the activition of NP- x B.AbstractObjective This study was to examine whether nuclear factor- c B(NFxB) was involved in the expression of monocyte chemoatractant protein-i (MCP- 1) in human mesangial cells induced by oxidized low-density lipoprotein(Ox-LDL),and to investigate the role of NF- K B and Ox-LDL in the pathogenesis of glomerulonephritis(GN).Methods (1) Human embryonic renal glomerular mesangial cells(MCs) was used as target cells.The effect of Ox-LDL on MCs Inhibitory K B a (I K B a) and MCP-1 protein level in both the presens and absense of NF- K B inhibitor pyrroledithiocarbomate(PDTC) was messured by cell ELISA.(2)Nuclear potein were extracted from quiescent MCs exposured with Ox-LDL or PDTC,then,activitis of transcriptional factors NF- K B were examined by electrophoresis mobility shift assay(EMSA).(3) Immunohistochemistry was used to observe the changes of p65 and MCP-i expression in MCs after Ox-LDL stimulation.(4) Immunohistochmitry was used to detect the expression of P65 ,I K B a and MCP-l in kidney biopsy specimens from patients with mesangial proliferative ON (MsPON) (n1 1), nonproliferative ON (nonPGN) (n8),and in structurally noumal kidneys (n5).
