Effect Of Zhen Qi Tong Mai Capsule On Aldose Reductase (AR) Gene MRNA Level Of Rat Retina Of Diabetic Retinopathy

Objective: To investigate the effect of Zhen Qi Tong Mai capsule ,Which had the fuctions of replenishing qi and nourishing yin, on aldose reductase (AR) mRNA level of DR rats retina and glucose. Methods: To establish the diabetic retinopathy animal model by an injection of streptozotocin(STZ). All DR rats treated by Zhen Qi Tong Mai after 3 months, which lasted for 3months. The level of AR mRNA of rat retina was analyzed by quantitatively RT- PCR method and evaluate the effect of Zhen Qi Tong Mai on AR mRNA level between groups. Results: 1. Zhen Qi Tong Mai capsule can obviously improve the DR rats' survival condition. 2. Three months later, DM rats displayed some early symptoms of diabetic retinopathy, Which means DR model established. 3. Compared with the model group, the content of glucose in Zhen Qi Tong Mai group were decreased evidently (P<0.01).4.The level of AR mRNA of rat retina is significantly different between diabetic retinopathy groups and normal controls, but there is no significant difference between the groups given Zhen Qi Tong Mai group. The level of AR mRNA of rat retina after injected STZ 6 months,is significantly higher than normal group (P<0.01).5.There is big significant difference between the groups given Zhen Qi Tong Mai group and diabetic retinopathy group. Zhen Qi Tong Mai can decrease the level of AR mRNA of rat retina (P<0.01). Conclusion: three months later, there are changes in retinal ultra structure in rats with diabetic Rats Zhen Qi Tong Mai capsule has the effect of decreasing glucose, and the level of AR mRNA of rat retina of diabetic retinopathy .Early treatment with Zhen Qi Tong Mai capsule may be helpful in delaying or possible inhibiting the underlying cause of diabetic retinopathy.
