Clinical Curative Effect Of Shenkangpian In Treating Patients Of Both Spleen Qi And Kidney Qi Deficiency Syndrome En Chronic Glomerulonephritis And The Effects On Expression Of IL-1 In Kidney Of The Adriamycin Nephropathy Rats

Objective: to analyze the adventage caused by shenkangpian on patients with chronic glomerulonephritis(CGN) and rats with adriamycin(ARD) nephropathy and to reveal the mechanism of it.Methods:clinic study:65 patients with CGN were divided into 2 groups:35 treated by shenkangpian and 30 treated by benazepril ,who were all treated for 8 weeks. Urinary protein excretion for 24 hours, red cell in urine, blood urea nitrogen(BUN), serum creatinine(Scr), creatinine clearance (Ccr), triglyceride(TG),cholesterol(CH).high density lipoprotein(HDL).low density lipoprotein(LDL),blood rheology and transforming growth interleukin-1 ( IL-1) in serum were tested before and after treatment. Experimental research:CGN was induced by intravenous injection of ADR. Rats were divided into 5 groups: untreated nephritic group. nephritic group treated by shenkangpian with low dosage,nephritic group treated by shenkangpian with high dosage,nephritic group treated by benazepril and normal control group.The groups were given drugs or water only for 6 weeks,respectively.Immunohistochemistry was used to examine the expression of type IV collagen(ColIV). fibronection(FN). lamin(LN) and IL-1.Proteinuria. Scr. BUN, TG CH, LDLand HDL were measured before and after treatment.Results: clinic study: the efficiency treated by shenkangpian and benazepril were 85.71% and 83.33%.respectively.Proteinuria ,red cells in urine.CH,HDL and LDL were improved by shenkangpian and benazepril with equal effect.The expression of IL-1 in serum was greatly decreased by shenkangpian. While shenkangpian was better in improving red cells in urine. Experimental research: compared with the untreated nephritic rats, proteinuria of rats treated by shenkangpian and benazepril was significantly reduced and albumin improved. Immunohistochemistry analysis showed shenkangpian and benazepril can decrease the expression of FN, LN, ColIV and IL-1. Besides, shenkangpian can decreased the TG of rats.Conclusions: to strengthen spleen and kidney,shenkangpian can ameliorate the syndromes of CGN,proteinuria,red cells in urine,lipid metabolism , IL-1 and extracellular matrix(ECM).So it can postpone the fibrosis of the kidney.
