Objectives: To investigate the influence of Combining RNAi-hTR and hTERT genes on telomerase activity of bladder cancer BIU-87 cell line and provide the new methods and evidence for RNAi in gene therapy of bladder cancer.Methods: 1.Two RNAi systems-PhTR-siRNA and PhTERT-siRNA targeting hTR and hTERT were constructed,screened and identified. 2.Biological effects to BIU-87 cells of respective or combining two RNAi systems, the growth inhibition of BIU-87 cells and telomerase activity were analysed. We measured the change of hTR and hTERT genes with RealTime-PCR.Results: The growth of bladder cancer BIU-87 cells were inhibited and telomerase activity was considerably decreased after respective or combining RNAi-hTR and hTERT genes. RealTime-PCR testified that HTR and hTERT genes was considerably decreased. The growth of bladder cancer BIU-87 cells were even more inhibited by combining RNAi than respective RNAi.Conclusions: Combining RNAi-hTR and hTERT genes can considerably inhibited the growth of bladder cancer BIU-87 cells.It may be one of the most promissing gene therapeutic strategies of bladder cancer.
