【Objective】To study the gene expression of the human gastric cancer with hepatic metastasis, investigate the differential gene expression in human gastric cancer with hepatic metastasis, discuss the probable mechanism of gastric cancer with hepatic metastasis.【Method】1.Collect 3 gastric cancer with hepatic metastasis patients'samples, including the primarily gastric cancer tissues and hepatic metastasis tissues , abstract and purify the total RNA from both of the two tissues, do the fluorescence labelling by respectively use the cy3,cy5 to obtain the probe of these two types of tissues by means of reverse transcription and hybridize with the array of SBC human 16kcDNA gene expression profile. Assess the differential expression gene by computer analysis and do the bioinformatics analysis for these acquired genes.2.Collect 5 gastric cancer with hepatic metastasis patients'samples, including the primarily gastric cancer tissues and hepatic metastasis tissues, abstract and purify the total RNA from both of the two tissues, reversely transcribe the first chain of cDNA, select the differential expression of cellular repressor of E1A-stimulated genes(CREG) and BRCA1-associated ring domain protein (BARD1), contrive the specific primer to authenticate with the first chain of cDNA by means of PR-PCR amplification.【Results】1.Screen the gene array and gain the differential expression genes. These genes'function refer to 11 kinds ,including anti-oxydize activity related gene, apoptosis related gene, cellular cohereing related gene, cellular metabolism related gene, cell cycle related gene and so on. Perform the comparing and cluster analysis of differrential expression gene between these arrays.2.Choose the manifestly up-regulating CREG gene and manifestly down-regulating BARD1 gene to contrive the specific primer to do the PT-PCR amplification with both of primarily gastric cancer tissue and hepatic metastasis tissue. These results display the same as the results of gene array.【Conclusions】Gene arrays high-flux choose hundreds of differential expression gene of gastric cancer with hepatic metastasis and choose CREG and BARD1 to disproof,in order to provide the evidences and references for the gene study of gastric cancer with hepatic metastasis.
