Background and Purpose The study is to explore the expression of the TNF-αand VEGF in the patients who suffer from the Intrahepatic Cholestasis of Pregnancy;to explore whether the up-regulation of caspases is correlated with the mechanism of Intrahepatic Cholestasis of Pregnancy (ICP).Methods 1.The ELISA methods were used to detect the TNF-αand VEGF expression in the peripheral blood, the blood in the Umbilical Vein and the amniotic fluid ,and the expression of TNF-αand VEGF of 27 ICP pregnant women and 30 normal pregnant women were to be compared.2.The TUNEL methods were used to detect the apoptosis index (AI) of the placenta tissues of the cases group (27 cases of ICP pregnant women) and the controls group (30 cases of normal pregnant women). The mmunochemistry methods were utilized to examine the expression of the Caspase-3 and Caspase-9 in the placenta tissues of ICP pregnant women and the normal pregnant women. Results 1.The level of the expression of TNF-αin the ICP patients'amniotic fluid and peripheral blood was lower than in that of the normal pregnant women and there is no significant difference between the expression of TNF-αin blood of Umbilical Vein between the two groups of women. The level of the expression of VEGF in the ICP patients'peripheral blood, Umbilical Vein blood and amniotic fluid was highter than in that of the control group (normal pregnant women).2.The apoptosis index in the syncytiotrophoblast of the ICP patients is higher than in the syncytiotrophoblast of the normal pregnant women (p<0.05). The expression of the Caspase-3 and Caspase-9 in syncytiotrophoblast of the ICP patients is higher than in the syncytiotrophoblast of the normal pregnant women (p<0.05 respectively).Conclusion 1.The lower level expression of TNF-αin the ICP patients'peripheral blood, amniotic fluid might demonstrate that TNF-αmainly acts on the trophoblastcyte of the placenta but not directly on the pregnant woman and the fetal of the ICP patient.2.When the fetal of the ICP patient becomes anoxic in the uterus, both pregnant woman and fetal might lead to up-regulation of VEGF, which might be to counterwork the emergency of anoxia. And the higher expression of VEGF is induced by endotheliocytes .3.The up-regulation of the apoptosis in the syncytiotrophoblast of the placenta is correlated with mechanism of ICP and it might play a role in the development of the ICP procedure.
