Objective: Intrahepatic cholestasis of pregnancy is a idiopathic diseaseduring pregnancy. Intrahepatic cholestasis of pregnancy increases the riskof fetal distress, preterm delivery, fetal loss and the morbidity andmortality of perinatal fetus. We hypothesize that the miR-155abnormalexpression associated with pathogenesis of ICP and adverse outcome.SO,the differential expression of miR-155in the placenta was comparedbetween normal pregnancy and pregnancy with ICP, to explore itsrelationship with intrahepatic cholestasis of pregnancy disease.Method: Ten cases of pregnancy undergoing cesarean delivery wasselected in Yongchuan District, Chongqing Maternal and Child HealthHospital from June2012to August2012. Among them,6cases of normalpregnancy were as control group and4cases of ICP pregnancy(tota bileacid>10umol/L) as experimental group.Tissue harvested trophoblastfrom under sterile conditions,Total RNA was extracted from liquidnitrogen grinding method. miR-155expression In each group was detectedby RT-PCR,The data were statistically analyzed by independent sample ttest. Result: Correction by internal standard GAPDH, the relativeexpression of miR-155was0.419±0.031in the6cases of normalpregnancy, and0.604±0.085in the4cases of normal pregnancy.Comparedwith the normal group, the relative expression of miR-155was significantlyincreased,the difference was statistically significant (P <0.05)Conclusion: miRNA genes differentially expressed in the placenta ofpatients with Intrahepatic cholestasis of pregnancy. miR-155up-regulatedin placental tissue of patients with ICP,should be involved in thepathogenesis of intrahepatic cholestasis of pregnancy.Genetic testing todetected the differential expression of miRNA in placental tissue of patientswith ICP, provides some basis study to further research on thepathogenesis of ICP and its adverse outcomes,and may provide us with apotential new diagnosis and treatment methods.
