Objective: To study the expression of Vascular Endothelial Growth Factor C(VEGF-C)and Vascular Endothelial Growth Factor D (VEGF-D)in laryngeal carcinoma and their relation with clinic pathologic features and lymphangiogensis .Methods:1.VEGF-C and VEGF-D expression were detected in 42 laryngeal cancer samples and 20 polyp of cord tissues by immunohistochemistry(IHC).Lymphangiogensis was evaluated by immunohistochemistry with the specific antibody D2-40.2. The expression of VEGF-C and VEGF-D protein was examined by western blot in 30 specimens of laryngeal carcinoma.Result:1.Result of IHC: The expression of VEGF-C,VEGF-D and MLVD was much higher in tumor tissue than in the polyp of cord tissue(P<0.05). Icreased expressions of VEGF-C and VEGF-D were significantly corelated with high T stage ,MLVD and lymph node metastasis (P<0.05)。But there was no significant association between their expression and age,grade of differation.The expression of VEGF-C was significantly correlated with the expression of VEGF-D (P<0.05).MLVD was significantly corelated with lymph node metastasis (P<0.05).2.Result of western blot: The expressions of VEGF-C and VEGF-D were higher in laryngeal cancer than those in adjacent normal tissue(P<0.01). Expressions of VEGF-C and VEGF-D were significantly corelated with high T stage and lymphatic metastasis(P<0.05)。The expression of VEGF-C was significantly correlated with the expression of VEGF-D (P<0.05).conclusion:1.Overexpression of VEGF-C and VEGF-D may play important roles in laryngeal tumorgenesis and correlated with T stage and regional lymph node metastasis.2.VEGF-C and VEGF-D may play important roles in lymphangiogenesis.3. MLVD was correlated with regional lymph node metastasis, which showed that lymphangiogenesis occurred in the development of laryngeal cancer.4.In laryngeal cancer the expression of VEGF-C was significantly correlated with the expression of VEGF-D .
