Objective: The laryngeal carcinoma is a very commontype of malignant tumors in otolaryngology, metastasis of cancer cells is a key prognostic factor for the patients's outcome. In past, being lack of lymphatic-specific markers,most studies about metastasis focused on the angiogenesis, whereas very few scholars paid attention to the lymphangiogenesis, the grow of new lymphatic vessels. Lymphatic vessel density (LVD) correlates with prognosis of patients's in some tumors. Vascular endothelial growth factor-D promotes lymphangiogenesis and angiogenesis in tumors and promotes the metastasis of cancer cells through lymphatic vessels.Endothelial Vascular Growth Factor (VEGF) also named Endothelial Permeability Growth Factor(VEPF),is an important positive regulator for angiogenesis ,and is one target of the anticancer research. Some nowadays studies suggest that the expression of Endothelial Vascular Growth Factor C (VEGF C) could stimulate lymphangiogenesis in tumors by activating its receptor or the lymphatic endothelial cells, then enhance the incidence of lymph node metastasis.This study leads to investigate the expression of VEGFR-3 and VEGF-C in laryngeal carcinoma and the relationship of them, with several clinical pathological characters and lymphatic vessel density (LVD) in laryngeal carcinoma, by by immunohistochemistry methods.Method: All the samples were collected from the patients admitted in the 1st affiliated Hospital of Chongqing Univercity of Medical Science, and confirmed by both clinical surgery and pathological examination. All specimen were embedded in paraffin and sectioned routinely, stained by immunohistochemical technique. LYVE-1 was used to mark lymphatic vessel endothelial. The expressions of VEGFR-3 and VEGF-C were observed under optical microscope.Results: The expression of VEGFR-3 and VEGF-C were higher in laryngeal carcinoma than those in vocal cord polyp ( P<0. 01). The expression of VEGFR-3 was significantly correlated with VEGF-C (r=0.575, P<0.01). The expressions of VEGFR-3 VEGF-C and LVD are associated positively, but have none relation with neither the tumors' type nor their T stage.Conclusion: The expression of VEGFR-3 may stimulate VEGF-C upregulation and the high expression of them may play an important role in lymphangiogenesis and the lymphatic spread in laryngeal tumors. Maybe some therapentic strategies will be found by restraining the expression.
