Objective: The ovary is an important organ in female reproductive system. The synthesizing and secreting of the gonadal hormone will decrease after the ovary aging , which lead to perimenopausal syndrome, osteoporosis, cardiovascular disease, obesity, diabetes and so on , it will impair women's physical and mental health. This research based on the region regulatory factors that influence ovary function and apoptosis of granular cell, study the mechanism of Gekko gecko against ovary aging through observing the affection of Gekko gecko ethanol extract on the expression of insulin-like growth factor-1(IGF-1), inhibin A(Inh A) and apoptosis of granular cells of the rats ovaries with different age, to find new methods and given theroy evidence to new medicine to delay ovary aging.Methods: 3 month(prepuberty) and 6 month (puberty)female rats were taken in study, 20 cases in each age ,rats of each age group were randomly divided into experimental group and control group,10 cases for each group. 2g(active compound).kg-1.d-1 of Gekko gecko ethanol extract was given to experimental group through intragastric administration for 15 days, corresponding volume of normal sodium was given to control group by the same way. Sacrifice rats after administration for 15 days , IGF-1 and Inh A of ovary paraffin section were detected by immunity fluorescence, apoptosis of granular cells were observed by TUNEL.Results: The expression of IGF-1 and the Inh A in experiment groups significantly raised and the apoptosis of granular cells significantly decreased compared with control groups (p<0.01) in both age group ; the expression of IGF-1 in 6 months age group was significantly higher than 3 months age group (p<0.05) in control groups; The expression of Inh A in 6 months of age group significantly higher than 3 months of age group (p<0.01) both in experiment groups and control groups.Conclusion: 1. Gekko gecko ethanol extract can significantly raise the expression of IGF-1 in rats ovaries, promoting the growth of follicles, it may be repress the apoptosis of granular cells and reduce follicular atresia through IGF-1 . 2. Gekko gecko ethanol extract can significantly raises the expression of Inh A in rats ovaries, thus improving ovary function , promoting the development of the dominant follicles and corpus luteum. This result discloses that Gekko gecko may improve ovulatory function of patients with anovulatory dysfunctional uterine bleeding, anovulatory infertility, and so on. 3. The contents of IGF-1 and Inh A are more affluent in ovary that has active function. 4. Gekko gecko ethanol extract can repress apoptosis of granular cells of rats ovaries obviously, and inhibit follicular atresia. This result discloses that Gekko gecko may delay ovary aging.
