Experimental study on the effect of kidney nourishing prescription onovarian morphology in rats with polycystic ovary syndrome. Purpose:To establish the pathological model of polycystic ovary rats and study the effect of kidney kidney and Resolving Phlegm Recipe on the morphology of rats.. Methods:Sixty 23-day-old female SD rats were randomly divided into 6 groups,The normal control group,model control group,drug treatment group,Chinese medicine high dose group, low dose of Chinese medicine group, combined treatment group,10 rats in each group.Using dehydroepiandrosterone(DHEA) method to establish the PCOS models. Then give the Bushenhuatanfang and(or) metformin to intervene,Chinese medicine high dose group. High dose group: treated with Bushenhuatanfang 1.8g/200g/d; low dose group were given Bushenhuatanfang 1.2g/200g/d; Drug treatment group: treated with metformin 0.008g/kg/d; combined treatment group: Bushenhuatanfang and Metformin(Bushenhuatanfang 1.8g/200g/d, Metformin 0.008g/kg/d), model control group: given volume physiological saline, continuous administration for 15 days.Study the differences of body weight, ovary index, uterus index, the number of ovarian follicle and corpus corpus, and the morphology of ovary of rats in each treatment group and normal control group. Results: 1. The changes of body weight, ovary index and uterus index of rats in each group Compared with the normal control group, the model group gained weight,(P<0.05); Ovarian weight increased(P<0.05), they are similiar to the most clinical PCOS patients which were overweight or obese, ovarian enlargement. Weight loss of rats in drug treatment group and combined treatment group, Compared with the normal control group(P>0.05), the results showed that there was a certain effects on the use of drugs; the experimental group were significantly lower than the model control group,the combination group was the best, and the normal control group had no significant difference, In addition to the model control group and low dose group, lower uterine weight, the weight of the rats in each group were restored to normal levels(compared with the normal group P>0.05). 2. The number of ovarian follicles and corpusCompared with the normal control group, the growth of the number of follicles increased, the number of mature follicles and corpus corpus decreased, there was significant difference(P<0.05), which suggested that the model was successful.Compared with the model control group,the experimental group can mature follicles and corpus luteum increased(P < 0.05), which combined treatment group curative effect best reach the level of the normal control group, low dose group and Western medicine group has increased, but there was no statistical significance, P > 0.05. 3. Morphological changes of ovarian tissue in rats of each groupPathological results showed that there were many corpus lutein and follicles in the ovary of the normal control group, the follicular cells and the corona were seen in the mature stage. Ovaries of the rats in model group visible multiple primary follicles and a small amount of secondary follicles, nearly mature follicles were not found in the oocytes, granulosa cell arranged loosely, to reduce the number of plies, partial loss, no corona radiata, theca cell layer is thicker. Compared with the model group, drug intervention of the four groups of rats ovarian pathology has been significantly improved, the ovaries were visible corpus luteum and multiple follicular, near mature stage follicles visible oocyte and corona radiata, granulosa cells arranged more closely, layers increased, theca cell layer is thin, high dose of Chinese medicine group and combined treatment group rats ovarian pathology improvement is particularly evident. Conclusion: 1. Through of PCOS model method of discussion and pre experiment several adequate evaluation model building, the final selection of dehydroepiandrosterone dehydroepiandrosterone(DHEA) induction in women with polycystic ovary syndrome model was established. The modeling method can be as an ideal PCOS animal model. 2. Bushenhuatanfang can improve the models weight, index of ovary, uterus index and ovarian follicle and corpus luteum number of polycystic ovary syndrome, the group with high dose of traditional Chinese medicine and combined treatment had the best effects. 3. The Bushenhuatanfang can obviously improve the morphological changes of ovary in rats with polycystic ovary syndrome, the high dose group of traditional Chinese medicine and the improvement of the pathological changes of the rats in the combined treatment group were more obvious.Experimental study on the effect of Bushenhuatanfang on femalehormone level in rats with polycystic ovary syndrome. Results: 1. Effect of Bushen Huatan Decoction on serum hormone levels in rats FSH, LHTest results show that: the normal control group compared with the control group, the serum FSH content, there was significant difference in LH(P<0.05), suggesting that the model was successful. After treatment, compared with normal control group, treatment group FSH and LH significantly decreased(P<0.05), no significant difference between each treatment group, combined treatment group of the two index decreased significantly.2. Effects of kidney nourishing prescription on serum hormone levels of T and Leptin in rats of each groupThe results showed that: compared with the normal control group, the model group T was significantly higher,(P<0.05); the combination treatment group T decreased significantly, compared with the normal control group no significant difference(P>0.05). Compared with the normal control group, except the combination group and leptin levels increased significantly(P < 0.01); with the model control group, the level of leptin in treatment group decreased significantly(P < 0.01), the combined treatment group of leptin level decreased significantly, and the normal group had no significant difference(P > 0.05). Conclusion: 1 after treatment, dehydroepiandrosterone(DHEA) polycystic ovary syndrome model induced by serum hormone levels in rats FSH, LH, T and Leptin were significantly improved. 2 the pharmacological mechanism of Bushen Huatan Decoction in the treatment of PCOS may be through the reduction of plasma T and LH levels, regulating the hypothalamic pituitary ovarian axis balance, promote ovulation and to achieve the goal of treatment. 3 this study shows that traditional Chinese medicine Bushen Huatan Decoction is effective in treating PCOS, metformin combined with Chinese medicine treatment group is better than that of pure Chinese medicine, traditional Chinese medicine and Western medicine for the treatment of PCOS is effective in treatment of promising.Experimental study on the effect of Bushenhuatanfang on expression of ovarian syndrome Insulin and its receptor on rat polycystic ovary Purpose:To investigate the effect of Bushenhuatanfang on polycystic ovary model rat ovarian insulin levels, and insulin receptor(insulin R), insulin-like growth factor-I(IGF-I), insulin-like growth factor-I receptor(IGF- I R), leptin(leptin) m RNA and protein expression level of traditional Chinese medicine and Western medicine and combined medication of PCOS, part of the mechanism of action. Materials and methods:Sixty 23-day-old female SD rats were randomly divided into 6 groups,The normal control group,model control group,drug treatment group,Chinese medicine high dose group, low dose of Chinese medicine group, combined treatment group,10 rats in each group.Using dehydroepiandrosterone(DHEA) method to establish the PCOS models. ELISA method was used to detect the serum insulin levels in rats of each group.The RT-PCR method for detection of rat ovarian tissue of insulin receptor, insulin R), insulin-like growth factor-I(IGF-I), insulin-like growth factor-I receptor(IGF- I R), leptin(leptin) m RNA expression levels.The Western blot was used to detect the rat ovarian tissue of insulin receptor, insulin R), insulin-like growth factor-I(IGF-I), insulin-like growth factor-I receptor(IGF- I R) and leptin(leptin) protein expression levels. Results: 1. Changes of serum insulin level and the expression of insulin receptor in rats of each group 1.1 Changes of serum insulin level in rats of each groupCompared With the normal control group compared the rest of the group the insulin content in serum increased significantly(P < 0.01); compared with the model group, compared the treatment group, the serum insulin decreased significantly(P < 0.01), the alliance treatment group decreased obviously. 1.2 The expression level of R Insulin in ovarian tissue of rats in each group 1.2.1The expression level of R m RNA Insulin in ovarian tissue of rats in each groupCompared with normal control group, compared the remaining rats ovarian tissue insulin receptor m RNA expression levels were significantly increased(P < 0.01); compared with the model group, high dose of Chinese medicine group, western medicine group, combination group, insulin receptor m RNA expression level was significantly decreased(P < 0.05), combination group decreased obviously(P < 0.01). 1.2.2Expression level of R Insulin protein in ovarian tissue of 1.2.2 ratsCompared with the normal control group compared the remaining rats ovarian tissue insulin receptor protein expression levels were significantly increased(P < 0.01); with the model group, in the treatment group, the insulin receptor protein expression levels decreased significantly(P < 0.01) in the combination group decreased significantly. 2. The expression level of IGF-I, IGF-IR, m RNA Leptin and protein in ovarian tissue of rats in each groupAnd compared to normal control group, model group of rat ovarian tissue IGF-I, IGF- I r, leptin m RNA and protein expression levels were significantly increased, and after drug treatment, compared with model group compared in the treatment group, the ovarian tissue IGF-I, IGF- I r, leptin m RNA and protein expression level decreased significantly, high dose of Chinese medicine group, western medicine group and combined treatment group, the change is most obvious, with significant difference(P < 0.05), combination group decreased significantly(P < 0.01). Conclusion: 1. After treatment, the level of insulin in the treatment group decreased significantly, traditional Chinese medicine high dose group and Western medicine group with similar levels, the best in the combination therapy group, suggesting that Bushen Huatan Decoction could significantly inhibit the polycystic ovary rat model serum insulin level. 2. Bushenhuatanfang can effectively reduce insulin R, IGF-I, IGF- I r, leptin m RNA and protein expression levels, and with traditional Chinese medicine and Western medicine combination therapy has better curative effect, indicating that Bushen Huatan Decoction Combined with metformin can effectively inhibit the polycystic ovary syndrome lesions in insulin resistance. 3. The treatment of polycystic ovary syndrome by means of Bushenhuatanfang may be achieved by regulating the level of insulin, decreasing the expression level of R Insulin, IGF-I, R IGF-I, m RNA Leptin and protein in ovarian tissue.
