OBJECT: To evaluate the effect of procyanidins in protecting lens against oxidative stress and to investigate the protection of procyanidins,to furnish the theoretic foundation and experimental evidence for further reaching on cataract drug therapy.METHODS: The healthy adult rabbit's lenses cultured in vitro were divided into 4 groups:Feton model group(oxidative stress group),groups treated with 100mg/L vitamin C , 100mg/L vitamin E, and with PC (50mg/L,100mg/L,200mg/L). After 24 hours of incubation,alteration of opacity was observed by photography and picture analysis; the histomorphology of lenses were examined under the light microscope; The levels of WSP,SOD,GSH-PX,CAT,T-AOC and MDA were tested.RESULT: 1,The transparency of all lenses were different.The figure gray scale was analyzed with Image Pro Plus 5.0 software, the relative gray scale in Feton model group and vitamin C group were lower than that in other groups (P<0.05),but the others were not different in statistics (P>0.05).2,Under the light microscope, Feton model group: lens anterior capsular membrane thickening, lens epithelial cells proliferation and arranged disorderly, cells waxed and nucleus appears multiformity. Lens cortex fiber layers arranged disorderly, much vesicle and water fissure among them, and big vesicle coalesced.Vitamin C group: lens anterior capsular membrane thickening too, basilar part of lens epithelial cells decoherenced generally from lens anterior capsular membrane. lens epithelial cells arranged loosely,layer derangement.There is a little cavitation among lens cortex fiber layers. Vitamin E group: lens anterior capsular membrane staining even and thin, complete and continuation, there was ves under lens anterior capsular membrane, cavitation among lens cortex fiber layers. Procyanidins groups: lens anterior capsular membrane continuation and smooth, lens epithelial cells tightly linked with lens anterior capsular membrane and lens cortex fiber,there was not vesicle under lens anterior capsular membrane and among lens epithelial cells.Lens epithelial cells arranged regularly, nucleus appeared unanimouse. Lens cortex fiber layers arranged tightly and regularly,no hollow and bubbic appeared.3,The level of WSP,GSH-PX and T-AOC in procyanidins groups were obviously higher than that of in Feton model group,vitamin C group and vitamin E group(P<0.01),but MDA lower than them (P<0.01). The content of SOD in procyanidins 100mg/L,200mg/L groups was much more higher than that of in the other three control groups(P<0.01),but that in procyanidins 50mg/L group was obviously higher than that of in Feton model group,vitamin C group too(P<0.01),and higher than vitamin E group(P<0.05). The level of CAT in procyanidins 200mg/L group was obviously higher than that of in Feton model group,vitamin C group(P<0.01),and procyanidins 100mg/L group higher than the two groups too(P<0.05),but the procyanidins 50mg/L group only higher than Feton model group(P<0.05).Compared with each other in procyanidins groups,we discovered that the level of SOD in 100mg/L,200mg/L was obviously higher than that of in 50mg/L group(P<0.01).In addition,the content of SOD,GSH-PX in vitamin E group was obviously higher than that of in Feton model group,vitamin C group (P<0.01),and T-AOC higher too (P<0.05), but MDA lower than Feton model group (P<0.05).There were no significant difference in all indexes between vitamin C group and Feton model group (P>0.05).CONCLUSION:1.Procyanidins can protect the lensof experimental rabbit against oxidative stress and maintain them diaphaneity.2.The level of WSP,SOD,GSH-PX ,CAT and T-AOC were increased,but MDA degraded. Procyanidins can inhibit obviously lipid peroxidation activity and decrease the cataract development.3. Vitamin C and vitamin E both appeared antioxidant ability in this experiment,but procyanidins had much more antioxidant activity than that of them.4.In this study, procyanidins can protect lens against oxidative stress displayed in cell biology and tissue. The toxic side effect of procyanidins has not been reported at present, therefore,this article can furnish the theoretic foundation and experimental evidence for further reaching on cataract drug therapy.
