Objective In this paper, the expression of HPV16/18E6,cyclinD1,hTERT were detected with immunohistochemical assays in nasopharyngeal carcinoma and laryngeal cancer. The correlation of the expression of the three genes and their clinicopathological features was investigated.Methods Immunohistochemical methods were used to detect the expression of HPV16/18E6,cyclinD1 and hTERT in paraffin-embedded specimens and the cells crawling the piece in 23 cases of nasopharyngeal carcinoma,42 cases of laryngeal cancer,two control groups .The results were compared.Results1. Positive expression of HPV16/18E6,cyclinD1 and hTERT were found in cell strains of HNE1 and Hep-2.2. The expression of HPV16/18E6,cyclinD1 and hTERT in NPC and its control group1) The expression of HPV16/18E6â‘ The positive rate of HPV16/18E6 protein in NPC was 26.1%(6/23), which was higher than in its control group , but there was no statistic difference(P>0.05).â‘¡The positive rate of HPV16/18E6 in NPC were not correlated with age, sex(P>0.05).â‘¢Expression of HPV16/18E6 in NPC were closely correlated with lymph node metastasis and clinical staging(P<0.05).2) The expression of cyclinD1â‘ The positive rate of cyclinD1 protein in NPC was 56.5%(13/23),which was higher than in its control group(P<0.01);â‘¡The positive rate of cyclinD1 in NPC were not correlated with age, sex and clinical staging(P>0.05).â‘¢The expression of cyclinD1 in NPC was closely correlated with lymph node metastasis(P<0.01).3) The expression of hTERTâ‘ The positive rate of hTERT protein in NPC was 82.6%(19/23),which was higher than in its control group(P<0.01);â‘¡The positive rate in NPC were not correlated with age, sex and clinical staging(P>0.05).â‘¢The expression of hTERT in NPC was closely correlated with lymph node metastasis(P<0.05).4) In NPC , correlation analysis revealed that there were positive relationship between the expression of HPV16/18E6 and cyclinD1,the expression of cyclinD1 and hTERT(P<0.05),the correlation between the expression of HPV16/18E6 and hTERT did not reach significance(P>0.05).3. The expression of HPV16/18E6,cyclinD1 and hTERT in laryngeal cancer and its control group 1) The positive rate of HPV16/18E6,cyclinD1 and hTERT protein in laryngeal cancer were 42.9%(18/42),50%(21/42) and 83.3%(35/42)respectively, which were higher than in its control group respectively(P<0.01).2) The expression of HPV16/18E6,cyclinD1,hTERT in laryngeal cancer were closely correlated with lymph node metastasis and clinical staging(P<0.05).3) Their positive rate in laryngeal cancer were not correlated with age, sex and clinical staging(P>0.05).4) Correlation analysis revealed that there were positive relationship between the expression of the three in laryngeal cancer(p<0.05).Conclusions HPV16/18 infection and overexpression of CyclinD1,hTERT may play an important role in carcinogenesis,development and metastasis in NPC and laryngeal cancer by activating together. They may be used as potential markers for early diagnosis,metastasis and prognosis in the two kinds of tumor.
