Objective: To find out the independent function of HGF/C-Met message pathway and VEGF-D message pathway and its reciprocity in the displaying invadation and angiogenesis of human squamous cell carcinoma of larynx, we use Immuno-histo-chemistry technique S-P method to test the expression of C-Met protein and VEGF-D in tumor and polyp of vocal cord as a control group .The expressions of Ki-67 ,D2-40and CD34 were also studied to deepen the understanding of bio-molecular mechanism of the two pathways .Providing some new views and probably some effective new method in the prevention diagnosis and treatment of carcinoma of squamous cell carcinoma of larynx.Methods: Forty two patients with the diagnosis of laryngeal SCC who were treated at the first affiliated hospital of CQMU ENT Department were included in the study. These patients were choosing randomly from a larger group of patients. 15 patients chosen had lymph nod metastases in the neck, and the remaining 28 patients did not. The mean age of these patients was 61.8 years old and 40 were male, two were female. None of the patient had undergone radiotherapy or chemotherapy before surgical excision. TNM classification was done according to UICC standard. (1992). Twenty patient suffered from polyp of vocal cord were also included as a control group. We investigated the expression of c-met and VEGF-D in paraffin-embedded tissue sections with immuno-histo-chemical staining S-P method for a relationship between the two factors and other histopathologic variables such as lymph node metastases, differentiation, micro-lymphatic vessel, micro-vessel, and invasion, Patten of growth. Use t test and sperman sas8.2 to analyze the results.Outcomes: The over expression of C-Met and VEGF-D were both found in LSCC tissues. Their level of expression is obviously higher than that of the control group and were found to be related with lymph node metastases, T grading and differentiation. Sex, age, location was not likely to affect their expression.Ki-67 positive cells and MV were found in experimental group and control group in different concentration. Whereas MLV were only found in LSCC tissues.There is a close relationship between C-Met and VEGF-D with MVD, MLVD, and Patten of growth of the tissue. The expression of C-Met protein and VEGF-D were also related.Conclusion: Over expression of C-Met protein in LSCC enhance the invasiveness and neck lymph node metastases, its possible mechanism maybe the promote angiogenesis and stimulate tumor cell growth.Over expression of VEGF-D in LSCC also promote angiogenesis and the Formation of lymph trouble.HGF/C-Met message pathway is closely related with VEGF-D message pathway and both can make tumor behave aggressively. Composed detection of C-Met protein and VEGF-D is helpful in diagnosis of neck lymph node metastases and preview the outcome.
