Purpose: to research association between bcl-2, p53, bax and neovascularization andsmoking, clinical pathology in 61 advanced squamous cell lung cancer, assess clinical value of bcl-2, p53, bax and micro-vessel density.Method: Squamous cell lung-cancer tissue samples from 61 patient who didn't treat before radiotherapy were assess by immunohistochemical method for expression of bcl-2, p53, bax, and the quantification of microvesel density (CD34). Assess che association between bcl-2, p53, bax and MVD and smoking, clinical pathology, analysis prognostic factor to influence patient survival. Results:1 .Correlation to oncoprotein and prognostic factors: bcl-2 positive expression was inversely associated with T category and clinical stage. MVD was correlated to T category and tissue differentiation. p53 expression was correlated to smoking index, The frequency of positive p53 increased with sm2.Association between oncoprotein bcl-2 was significantly associated with MVD. There was inverse correlation between p53 and bax expression.3.Univariate analysis of the patients population at large show T category, N category, clinical stage, tissue differentiation, bcl-2 and MVD were predictive for overall survival. In multivariate analysis, only T category and bcl-2 had independent prognostic influence on overall survival. Bcl-2 belong to protective factor,conclusion1. bcl-2 was a independent prognostic factor, and was protective factor. Bcl-2 expression could depress angiogenesis and was helpful to choosing therapy method.2. Angiogenesis prompted tumor cell to proliferate, depression to Angiogenesis would become a method to cure lung cancer.3. smoking can lead to p53 gene mutation ,so smoking was a dangerous factor to lung cancer.
