Objective:To investigate the association between rs7903146 Polymorphism in the transcription factor 7-like 2 gene and type 2 diabetes mellitus(T2DM) in Chinese Han nationality in Chengdu area.Research design and methods:There are 3 groups in our research which included(1)Group of type 2 diabetic pedigrees:Consisting of 37 male and 37 female patients.9 pedigrees of T2DM(n=74) were recruited in Chengdu area,which included 29 T2DM patients,15 patients with impaired glucose tolerence (IGT) and 30 individuals with normal glucose tolerance (NGT);(2)Group of sporadic T2DM:92 T2DM patients without diabetes family history were recruited,including 45 male and 47 female(mean age 60.61±10.24).(3)group of normol control subjects who have no history of diabetes mellitus were confirmed being normal glucose tolerance by OGTT or with FBG<5.6mmol/L plus HbA1c<6.2%, included 31 male and 49 female,with mean age58.3±15.96. The genotypes of rs7903146 Polymorphism in the TCF7L2 gene of all 246 subjects were analyzed by RT-PCR. clinical characteristics,such as BMI, blood pressure, the plasma triglyceride(TG),total cholesterol(TC) and high density lipoprotein cholesterol(HDL-C)level were measured in all subjects.Results:1.The genotype distribution of rs7903146 Polymorphism in the TCF7L2 gene were in Hardy-Weinberg equilibrium (X~2=0.243,P=0.62).2.The frequency of genotypes in group of T2DM patients of type 2 diabetic pedigrees(P-Y2DM) were C/C89.6%,C/T 10.4%respectively, in group of IGT patients of type 2 diabetic pedigrees(P-IGT) were C/C93.3%,C/T6.7%and in group of normal glucose tolerance subjects of type 2 diabetic pedigrees(P-NGT) were C/C96.7%,C/T3.3%.While,The frequency of T allele were 5.2%,3.3%and 1.67 respectively.In Chi-square Test showed,there are no significance in genotype frequency and allele frequency among subgroups in T2DM pedigrees.3The C/C and C/T genotype frequency in PT2DM were 89.6%and 10.4%respectively, in group of sporadic T2DM(ST2DM) were 91.3%and 8.7%,and in group of normol control subjects(NC)were 97.5%and 2.5%.While,The frequency of T allele were 5.2%,4.3%,1.3%respectively. In Chi-square Test showed,there are no significance in genotype frequency and allele frequency.4.The logistic regression analysis demonstrates that OR of C/T genotype are no significance.In P-T2DM&P-IGT and PNGT OR is 0.367, 95%CI=0.042-3.2,and in ST2DM and NC OR is0.288, 95%CI=0.063-1.315. Also the logistic regression analysis shows that OR of T allele are no significance. In P-T2DM&P-IGT and P-NGT OR is0.367, 95%CI=0.042-3.2 and in ST2DM and NC OR is0.302, 95%CI=0.043-2.1.Although there were many studies had indicated that the polymorphism of rs7903146 of TCF7L2 was one of the risk factor associated with T2DM in white population.Our study showed that there was no evidence to consider the polymorphism of rs7903146 of TCF7L2 as the main risk gene of T2DM in china Han nationality in Chengdu area. Conclusion: rs7903146 Polymorphism in the transcription factor 7-like 2 gene does not seemingly play a role in type 2 diabetes of chinese.
