streptococcus mutans are the bacteria most commonly associated with the human dental carries. A major virulence attribute of these and other cariogenic bacteria is acid tolerance. The acid tolerance mechanism, including the F-ATPase, have begun to be investigated in detail. The study also including that carries are prevented and cured by traditional Chinese herbs. Many crude extract of Chinese drug were found effective in antibiosis. In a word , the study on enzyme including F-ATPase related caries is important to finding new treatment of dental caries. (1) we interfered the culture of streptococcus mutans ATCC25175 in drugs of different level, get the right level affect the bacteria most. (2) the culture of streptococcus mutans ATCC25175 were subjected to cycles of freezing and thawing and then separately resuspended in Tris-HcL buffer at desired interfered drug level. After reactioned with the ATP, the activity of Proton translocating membrane ATPase were aasayed. It has obviously inhibited effects by using fluoride and crude extract of Chinese drug, while these drugs of different level don't have obvious effets on the virulence factor. The influential factor in every group don't compared because of different drug level. In a word, Just like fluoride, crude extract of Chinese drugs have intensive inhibitive effects on the ATPase activity. This study suggest: application of the effective ways to prevent caries, and it also plays an important role in regulating the ecological balance of dental plaque.
