Objective: The purpose of this study is①to learn the effects of ginseng on the life-span and the lipid per-oxidation of Drosophila Melanogaster②to discuss the protected effect and the mechanism of ginseng about the damage of the lipid per-oxidation result by heated cooking oil on Drosophila③to study the impact of ginseng on the gene expression of Cu, Zn-SOD,Mn-SOD and CAT④to determine the different extraction process of saponins from ginsenosid. Methods:①Longevity test was carried out to find the influence of Ginseng on Drosophila's life-span. We detected the activity of total-SOD and the concentration of MDA by biochemical analysis methods, in order to research the relation between effects of Ginseng on the antioxidant system and aging in Drosophila.②Longevity test and biochemical analysis methods were used to discuss the protected effect and the mechanism of ginseng about the damage of the lipid per-oxidation result by used cooking oil in Drosophila.③RT-PCR was an effective technique to display the alterations in the gene expression of Cu, Zn-SOD,Mn-SOD and CAT in Drosophila's head after Ginseng exposure.④The determination of Ginsensides of different extraction process was studied by High performance liquid chromatography(HPLC) Results:①Every test groups MLS and MMLS are longer than the control group. The activity of SOD is increased along with the increasing the concentration of ginseng; the concentration of MDA is decreased along with the increasing the concentration of ginseng②the MLS and MMLS of the group faded by high concentration ginseng and heated cooking oil are higher than the groups faded by cooking oil, and lower than the control group;the activity of SOD of Drosophila faded by high concentration ginseng and heated cooking oil are higher than the groups faded by cooking oil; the concentration of MDA of Drosophila faded by high concentration ginseng and heated cooking oil are lower than the groups faded by cooking oil③some certain concentration of ginseng can increase the gene expresion of Cu, Zn-SOD, Mn-SOD and CAT.④the linear range of ginsenoside Rbl, Re and Rgl was 0.5003μtg~7.5045μg, 0.5009μg~7.5135μg and 0.5012μg~7.518μg, respectly. Conclusion:①ginseng can increase the activity of SOD, decrease the concentration of MOD and prolong the life-span of Drosophila Melanogaster②the damage of the lipid per-oxidation result by heated cooking oil in Drosophila can be resisted in a certain degree by some concentrations of ginseng③ginseng can increase the gene expression of Cu, Zn-SOD,Mn-SOD and CAT④the most effective extraction processes is SUO methed, and ultrasonic method is better than warm soaking method.Key Words: Panax Ginseng; Gingsenoside; Longevity test; Drosophila Melanogaster; antioxidase; RT-PCR; HPLC; extraction process; used cooking oil...
