| Objective To investigate the effects of lanthanum chloride on serum inflammation media in diabetic rats.Methods Blood was sampled from the orbit veins with fasting abdomen in the morning to measure the serum insulin,C peptide,interleukin-6 and tumour necrosis factor-αin 50 healthy Male Sprague Dawley rats. Blood glucose levels of rat caudal veins were measured by glucometer Surestep at the same time.Inject the left lower quadrant of rats with 1% streptozotocin by weight and measure the blood glucose levels of rat caudal veins by glucometer Surestep after 7 days.Regard the rats with blood glucose level≥13.9mmol/L as the model of diabetes mellitus.Diabetic rats were randomly divided into four groups:low-dose,middle-dose,high-dose and control groups. Inject the left lower quadrant of diabetic rats with low-dose(0.01mg/kg),middle-dose(0.05mg/kg),high-dose(0.25mg/kg) lanthanum chloride and the same volume of saline respectively every other day for 8 weeks. All the rats were allowed to drink and eat freely. Fasting and random blood glucose was measured at 0th week,4th week and 8th week.Fasting serum insulin,C peptide,interleukin-6 and tumour necrosis factor-αfrom the orbit vein was measured by radioimmunoassay at 0th week and 8th week.All the data were expressed in X±s,and analyzed with analysis of variance and canonical correlation analysis by SPSS 11.5 for windows.Results The level of random blood glucose was significantly decreased, that of fasting serum insulin,C peptide was elevated and that of fasting serum interleukin-6 and tumour necrosis factor-αwas decreased in low-dose group after 8 weeks. The differences were statistically significant compared with the other groups(P<0.01); There was a positive correlation between the level of fasting serum interleukin-6 and tumour necrosis factor-αand the level of random blood glucose,and a negative correlation between the level of fasting serum interleukin-6 and tumour necrosis factor-αand the level of fasting serum insulin,C peptide.Conclusion Low-dose lanthanum chloride can inhibit the effect of serum inflammation media,decrease the apoptosis of pancreatic isletβcells,increase the level of fasting serum insulin and decrease the level of blood glucose effectively. |
