Objective:Since rare earth exposure can have unnecessary effects on the environment and human body,this study explored the biological effects and toxicity comparison of rare earth element lanthanum on the first and second generations of C57 mice from multiple dimensions.First,it can enrich the toxicity evaluation of rare earth element lanthanum and provide theoretical basis and data support for the safety evaluation of lanthanum;Second,through comparison,we can reveal whether the influence of rare earth element La on the offspring will be greater with the increase of generation generation.These data are of reference significance for the groups who have long-term exposure to rare earth element La to take effective measures.Methods:Lanthanum chloride was prepared into 0 mg/kg,10 mg/kg,20 mg/kg and 40 mg/kg solutions,and was named as control group(Con),low dose(LD),medium dose(MD)and high dose(HD).Acquisition and toxicity of F1:16 specific non-pathogen(SPF)C57BL/6j female mice(F0,7-8 weeks old)were randomly divided into four groups,four in each group,and paired with male mice in the ratio of 2:1.The parents drank freely before and after pregnancy according to the corresponding dose of lanthanum chloride solution.F1 was ingested lanthanum by the mother before weaning,and was ingested by free drinking water for 3 months after weaning.Acquisition and toxicity of F2:two males and four females were randomly selected from F1 generation mice(7-8 weeks)of each dose group for mating.Follow the dose and method corresponding to F1 for 3 months.The biological effects of lanthanum on C57 mice were evaluated by studying the effect indexes such as the content of La in each organ,body weight and organ weight,oxidative stress,lipid metabolism,liver histopathology and transaminase activity,and using the methods of t-test,one-way ANOVA and LSD test to statistically analyze the relevant quantitative data.Results:This study explored the effects of lanthanum chloride exposure on organ weight,oxidative stress level,lipid metabolism level,transaminase activity and liver histomorphology of C57 mice from multiple dimensions.The specific results are as follows:1.Lanthanum can be accumulated in various organs,and its main accumulated organs are liver,lung,epididymis and kidney,which will affect the development of mouse organs.With the increase of algebra,the distribution of lanthanum in the body will change,and the influence on liver and kidney will be greater;2.Lanthanum exposure can increase the MDA level of mice,resulting in lipid peroxidation of the body,but it does not exceed the compensatory capacity of the body,and has less impact on the offspring;3.Lanthanum exposure can affect the activity of transaminase and the pathological structure of liver tissue in mice,thus leading to the decline of liver function,and then affect the level of lipid metabolism,and the influence on F2 is less.Conclusion:Under the conditions of this experiment,it can be considered that long-term exposure to lanthanum has a certain subchronic toxic effect on oxidative stress level,lipid metabolism level,transaminase activity and liver histomorphology of mice,and its effect will weaken with the increase of generations.
