Objective: To observe the acute and chronic effect of human recombination leptin on humen lipocyte metoblism, and explore the regulation of leptin on lipocyte lipopexia.Method: Get human subcutaneous adipose tissue, extract and culture preadipocyte. Induce preadipoctyes differentiating toward adipocytes after fusion of the preadipocytes. After been proved to be adipocyte by observing under microscope and stained with rathonum red O, divined adipocytes into 4 groups by random: control group,lower concentration group,common concentration group and higher concentration group, in which the leptin,s concentration was 0ng/ml, 10ng/ml, 100ng/ml, 1000ng/ml each. Measure the concentration of free fatty acid and glycerol in nutrient medium and detect integral lipochondrias optical density in the adipocytes at the time of 4h, 48h, 72h. Analyze the data and observe the acute and chronic effecet of human recombination leptin on human adipocyte katabolism.Result: 1,At the time of 4 hour , the concentration of free fatty acid and glycerol in high leptin concentration group increased by 312%(p<0.01) and 17%(p<0.01) about respective contrast to the control group; the integral lipochondrias optical density decreased by 35%(p<0.01) about. The concentration of free fatty acid and glycerol in low leptin concentration group increased by 87%(p<0.01) and 5%(p<0.01) about respective; the integral lipochondrias optical density decreased by 12%(p<0.01) about. The concentration of free fatty acid and glycerol in common leptin concentration group increased by 181%(p<0.01) and 8%(p<0.01) about respective; the integral lipochondrias optical density decreased by 21% about. The concengtration of free fatty acid and glycerol had positive correlation with the concentration of leptin , the integral lipochondrias optical density had negative correlation with the concentration of leptin 2. At the time of 48h, the concentration of free fatty acid and glycerol in commom leptin concentration group increased by 334%(p<0.01) and 31%(p<0.01) about respective; the integral lipochondrias optical density decreased by 46%(p<0.01) about. The concentration of free fatty acid and glycerol in low leptin concentration group increased by 94% and 16%(p<0.01) about respective; the integral lipochondrias optical density decreased by 18%(p<0.01) about. the concentration of free fatty acid and glycerol in high leptin concentration group increased by 252%(p<0.01) and 22%(p<0.01) about respective; the integral lipochondrias optical density decreased by 31%(p<0.01) about. The concengtration of free fatty acid and glycerol and the integral lipochondrias optical density had no correlation with the concentration of the leptin.3. At the time of 72 hour, the concentration of free fatty acid and glycerol in common leptin concentration group increased by 382%(p<0.01) and 36%(p<0.01) about respective. The integral lipochondrias optical density decreased by 49%(p<0.01) about. the concentration of free fatty acid and glycerol in low leptin concentration group increased by 103%(p<0.01) and 13%(p<0.01) about respective; the integral lipochondrias optical density decreased by 19%(p<0.01) about. the concentration of free fatty acid and glycerol in high leptin concentration group increased by 259%(p<0.01) and 24%(p<0.01) about respective; the integral lipochondrias optical density decreased by 43% (p<0.01)about. The concengtration of free fatty acid and glycerol and the integral lipochondrias optical density had no correlation with the concentration of the leptin.4. At the time of 4 hour, the does dependent was observed about the leptin,s effect on adipocyte, and the leptin of 1000ng/ml had the maximal effect;the time dependent was not observed during the chronic effect of leptin on human adipocyte. Conclusion: The increase of katabolism and decrease of lipopexia were observed during both the acute and chronic act of human recombination leptin on human adipoctye. And the acute act of leptin had the feature of dose dependent, but the chronic one had not. The time dependent was not observed the chronic regulation of leptin to metabolism and lipopexia.
