Gene Transfer Into The Mammalian Inner Ear Using A RNAi Non-viral Vector

Objective To determine the possibility of a locally used RNAi non-viral vector in the living mammalian inner ear.Methods The vector mixed with cationic polymer was infused through the round window, the expression measured by immunofluorescence, DPOAE and ABR recorded 1d 3d 7d after infusion separately.Results The expression of the vector was detected 1d after the infusion and still remained 7d post-infusion. And the hearing loss was temporary 1 day post-infusion and eliminated afterward.Conclusion The RNAi non-viral vector can express in the inner ear harmlessly and is probably a promising tool for the down regulation gene therapy in the future.
