Objective:Study the expression and it's significance of P27 and p-ERK1/2 proteins in human gastric carcinoma,and investigate the possible relationship between them.Methods:P27 and p-ERK1/2 proteins expressions were examined by immunohisto chemical s-p staining method in 46 cases of gastric carcinoma and 10 cases of human normal tissue using P27 and p-ERK1/2 monoclonal antibodys.Result:(1)Positive expression rate of P27 in gastric carcinoma was 28.3% (13/46),which was significantly lower than that in normal tissues(90%,P<0.01). The expression level of P27 was related to histological differentiation, extent of infiltrating , lymphnode metastasis(P<0.05).(2)Positive expression rate of p-ERK1/2 in gastric carcinoma was 47.8%(22/46),which was significantly higher than that in normal tissues(10%, P<0.05). The expression level of p-ERK1/2 was related to extent of infiltrating , lymphnode metastasis and TNM stage(P<0.05).(3)There is a significant inverse correlation in the expression of P27 and p-ERK1/2.(P<0.05).Conclusion:The overexpression of p-ERK1/2 and hypoexpression of P27 were found in gastric carcinoma; the expression of P27 and p-ERK1/2 protein were strongly related to the infiltration and mestastasis of gastric carcinoma;the expression of P27 was negative correlation with p-ERK1/2 rankly. The activity of p-ERK1/2 can regulate the expression of P27 and both of them may play an important role in the genesis and development of human gastric carcinoma. It benefits to the early diagnosis and prognosis of cancer, even to the strategy selection in treatment.
