Objective To observe the infection of Helicobacter pylori(Hp)in chronic superficial gastritis,chronic atrophic gastritis,intestinal metaplasia,intraepithelial neoplasia,early gastric cancer and advanced gastric cancer,and the activation of phosphorylated extracellular signal-regulated kinase 1/2 Related proteins c-myc,c-Fos,ATF4 expression characteristics of gastric cancer during the development of extracellular signal-regulated kinase and its mechanism.Methods Immunohistochemical staining was used to detect chronic superficial gastritis(20 cases),chronic atrophic gastritis(20 cases),intestinal metaplasia(19 cases),low grade intraepithelial neoplasia(20 cases),high grade intraepithelial neoplasia The expression of P-ERK1/2,ERK1/2,c-myc,c-Fos and ATF4 in early gastric cancer(31 cases)and advanced gastric cancer(73 cases)Semiquantitative evaluation of the expression of P-ERK1/2,ERK1/2,c-myc,c-Fos,ATF4 in each group.Results In each group of immunohistochemistry result show: The expression of HP in high magnification in gastric pits or glandular cavity visible brown short rod,comma shaped or spherical.In chronic superficial gastritis(55%),intestinal metaplasia(47.4%),high grade intraepithelial neoplasia groups(41.7%),early gastric cancer(16.1%)and advanced gastric carcinoma(8.2%),the infection rate of Hp was lower than the chronic atrophic gastritis(94.7%)(P<0.05).The expression of P-ERK1/2 and ERK1/2 was positive in cytoplasm or membrane,and the expre-ssion of P-ERK1/2 in advanced gastric cancer was higher than that in mild chronic gastritis(15.0%)and intestinal metaplasia(15.8%(P<0.05).The expression of P-ERK1/2 in early gastric cancer was higher than that in chronic gastritis and chronic atrophic gastritis(35.0%)And intestinal metaplasia(P<0.05).c-myc was located in the nucleus.The expression of c-myc in advanced gastric cancer(34.2%)was lower than that in low-grade intraepithelial neoplasia(60.0)(P<0.05).c-Fos positive cells located in the cytoplasm and membrane,advanced gastric cancer(83.6%)c-Fos expression rate was higher than low-grade intraepithelial neoplasia(35.0%),chronic superficial gastritis(50.0%),chronic atrophic gastritis(26.3%)(P<0.05).The expression of c-Fos in early gastric cancer(67.7%)and high-grade intraepithelial neoplasia(75.0%)was higher than that in low-grade intraepithelial neoplasia and atrophic gastritis(P<0.05).The positive rates of P-ERK1/2,ERK1/2,ATF-4,c-myc and c-Fos in gastric cancer were statistically significant compared with non-cancer group(P<0.05).Compared with Hp infection group and Hp noninfection group,the expression rates of P-ERK1/2,ERK1/2,c-myc and c-Fos in Hp-positive group were higher than those in Hp-negative group,Hp-positive P-ERK1/2 was higher in the low-grade intraepithelial neoplasia than in the Hp negative group,and the AFP4 in the Hp-positive group was higher in the advanced gastric cancer group than in the Hp negative group(P<0.05).Conclusion Hp infection is related to the expression of ERK1/2,P-ERK1/2,c-myc,c-Fos and ATF-4 during the development of gastric cancer.
