Objective: Nuclear factor kB (NF-kB) is a nuclear transcription factor that regulates expression of a large number of genes.The current study was to investigate the active- ty change and significance of NF- B p65 in mice with peripheral tolerance induced by donor specific spleen cell transfusion (DST).Methods: BALB/c to C3H skin allograft model was employed in all groups.Skin and spleen were collected on the seventh day from skin transplantation C3H mice which intravenous injected 1×108 spleen cells of BALB/c for 3 or 7days before trans- plantation. The immune function was detected by splenic lymphocyte proliferation test and mean survival time(MST) of skin allograft. The activity of NF- B p65 was examined by immunohistochemical staining.Result: Contrast with comparison group, DST for 7 days before skin transplantation can prolong the mean survival time(MST) of skin allograft and suppress the antigen- specific lymphocyte proliferation reaction; the activity of NF- B p65 in skin allograft was increased notably; however, the activity of NF- B p65 in spleen was decreased. Conclusion: The peripheral tolerance induced by DST may be related to the change of NF- B p65 activity in different places.
