Object: To study the effect of benfotiamine on proliferation and apoptosis of bovine retinal capillary endothelial cells cultured in high glucose.Methods: Bovine retinal capillary endothelial cells were cultured in high (30 mmol/L) glucose with different concentration of benfotiamine ( 0,25μmol/L,50μmol/L,75μmol/L,100μmol/L) in vitro . 1. MTT assay were used to observe the effect of benfotiamine on the cells proliferation when they were separately cultured in 37℃for 24h,48h and 72h, analysed the data and chose a proper medicine concentration. 2. Cells were cultured in high glucose with chosen concentration of benfotiamine for 48h. Flowcytometry was used to analyze the cells apoptosis .Result: Treated with 75μmol/L benfotiamine, the value of OD (MTT) ˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉˉincreased obviously, compared with that of the control group the difference was significance (p<0.01) and the OD value was increased in a dose dependent manner. Flowcytometry showed that the endothelial cells cultured in high glucose with 75μmol/L benfotiamine had lower rate of apoptosis.Conclution: Certain concention of benfotiamine (75μmol/L) can protect bovine retinal capillary endothelial cells cultured in high glucose, maybe due to it can reduce the apoptosis of the retinal capillary endothelial cells. Thus it may be a potential medicine to prevent diabetic capillary complications.
