Objective: To explore the effects of different concentrations of procyanidin(procyanidin,PC)on the proliferation of human retinal vascular endothelial cells(Human retinal vascular endothelial cells,HRCECs)and the expression of VEGF(vascular endothelial growth factor,VEGF).Method:1.Three to four generations of primary culture HRCECs were taken as the study objects.The experiment was divided into six groups: low sugar control group(TL),high sugar control group(TH),high sugar + 200 ?g / ml procyanidin group(T1),high sugar + 400 ?g / ml procyanidingroup(T2),high sugar + 600 ?g / ml procyanidingroup(T3),high sugar + 800 ?g / ml procyanidingroup(T4).After adding these concentrations of procyanidinon,HRCECs were cultured from 24 hours,48 hours,72 hours.2.MTT method was used to detect the proliferation of HRCECs,Western blot(WB)was used to detect the expression of VEGF in HRCECs.Results: 1.Compared with the low glucose control group,the MTT absorbance of HRCECs in the high glucose control group increased significantly after cultivating 24 hours,48 hours and 72 hours(P< 0.05);the VEGF expression in TH group was higher and the difference was statistically significant(P< 0.05).2.After 24 hours,48 hours and 72 hours of incubation with different concentrations of proanthocyanidins,the MTT absorption values of T1,T2,T3 and T4 groups were significantly lower than TH group(P< 0.05).At the same time,the higher the drug concentration,the absorption value of MTT was smaller and smaller,the difference is statistically significant(P< 0.05).3.After 24 hours,48 hours and 72 hours incubation with different concentrations of proanthocyanidins,compared with TH group,the expression of VEGF decreased significantly(P< 0.05).AT the same action time,the higher the drug concentration is,the smaller the gray value is.Conclusion: 1.In high glucose environment,procyanidin could inhibit the proliferation of HRCECs in vitro in a concentration dependent manner.2.In high glucose environment,procyanidin could inhibit the expression of VEGF in cultured HRCECs,which shows a certain concertration dependence.Procyanidin maybe could inhibit the formation of retinal neovascularization.
