Objective1. To investigate the optimal compatibility ratio and chemical basis of Rhizoma Coptidis and dried Rehmannia root .2. To explore the relationship of"compatibility-material basis-pharmacological effect''and elucidate the scientific principle of formula compatibility in Huanglian Bolus . MethodPart one Research on pharmaceutical effect of formula compatibility of Huanglian Bolus——Influence on Huanglian Bolus's pharmaceutical effects of streptozotocin–induced diabetic rats of different compatibility ratio of Rhizoma Coptidis and dried Rehmannia rootThe murine diabetes models were induced by tail intravenous injection of streptozotocin ( STZ , 40 mg·kg-1 ) , then modeled diabetic animals were randomly divided into control group,Huanglian (RC) group,Shengdi (DR) group,RC:DR 1:1 group,RC:DR 1:4 group,RC:DR 1:8 group with normal rats as normal control . After the intragastric administration for 14 days, the indices such as body weight fasting blood glucose ( FBG ),total cholesterol ( TC ),triglycerides ( TG ) and detected pancreatic pathological morphology were observed between each group.Part two Research on material basis of compatibility of Huanglian Bolus——Influence on dissolvability of berberine and cataplol as effective contents of Huanglian Bolus of different compatibility ratio of Rhizoma Coptidis and dried Rehmannia root1. Influence on content of berberine in Huanglian Bolus with different compatibility ratio of Rhizoma Coptidis and dried Rehmannia root The method of determining the berberine of herbal decoction was established by high-performance liquid chromatography (HPLC ) , the accuracy,repetition,stability and recovery detections were reviewed in five samples regarding RC:DR 1:1 as standard .2. Influence on content of catalpol in Huanglian Bolus with different compatibility ratio of Rhizoma Coptidis and dried Rehmannia root The method of determining the catalpol of herbal decoction was established by HPLC , the repetition and content of five detections were reviewed in five samples regarding RC:DR 1:1 as standard .ResultsPart One Analysis of influence on Huanglian Bolus's pharmaceutical effects of streptozotocin–induced diabetic rats of different compatibility ratio of Rhizoma Coptidis and dried Rehmannia root1. Body weight change before and after treatment The treatment groups and model group were significantly lighter than normal group ( P<0.05 ) before treatment . After treatment , treatment group gained more weight than model one ( P<0.05 ) . Huanglian group and RC:DR 1:8 made no significant difference with normal one ( P<0.05 ) .2. Hypoglycemic effect of Huanglian Bolus on diabetic rat models Before treatment , FBG of model group or treatment group were much higher than normal one ( P<0.01 ) , but there was no significant difference between modeled rats ; After treatment , the treatment groups demonstrated lower FBG than model one ( P<0.01 ) , Huanglian group,RC:DR 1:1 and RC:DR 1:4 had no significant difference with each other , but they had significantly lower FBG than Shengdi group ( P<0.01 ) and had higher FBG than RC: DR 1:8 ( P<0.01 ) , FBG of RC:DR 1:8 had lower than other modeled rats ( P<0.01 ).3. Lipids change of Huanglian Bolus on diabetic rat models Influence on TC after treatment : All treatment are much lower TC than model one ( P<0.05 ) ; Shengdi,RC:DR 1:4,RC:DR 1:8 made no different TC with normal one , Huanglian group and RC:DR 1:1 showed lower TC than normal one ( P<0.05 ) . Shengdi,RC:DR 1:4 and RC:DR 1:8 had no different TC with each other , but they had lower TC ( P<0.05 ) than Huanglian group and RC:DR 1:1 which didn't have different TC with each other . Influence on TG after treatment : Each treatment group had significantly lower TG than model one ( P<0.05 ); treatment group except RC:DR 1:8 had higher TG than normal one ( P<0.05 ) ; No significant difference of TG were observed between Shengdi,RC:DR 1:1 and RC:DR 1:4 , but they had lower TG than Huanglian group ( P<0.05 ) and had higher TG than RC:DR 1:8 ( P<0.05 ) ; RC:DR 1:8 had distinctive lower TG than other treatment group .4. pathological paraffin observation between groups Pancreatic pathological paraffin section showed: the islet of normal group had clear boundary with rich numerous cells , the islet of model group had blurred boundary and disturbed form of bloated , hazy islet cell. Islet in Huanglian group and RC:DR 1:8 took similar configuration while Shengdi,RC:DR 1:1 and 1:4 group demonstrated badly-ruined islet which was still better than model one.Part Two Influence on dissolvability of berberine and cataplol as effective contents of Huanglian Bolus of different compatibility ratio of Rhizoma Coptidis and dried Rehmannia root1. Influence on dissolvability of berberine in Huanglian Bolus of different compatibility ratio of Rhizoma Coptidis and dried Rehmannia root It was a effective,stable,reliable method to detect the content of berberine. The accuracy ( 1.6 % ),repetition( 1.29 %),stability ( 2.58 % ),recovery ( 2.31 %,1.18 %,1.72 % ) were invariable. The dissolvability ratio of DR : RC 1: 0,0 : 1,1: 1,1: 4,1: 8 were respectively 2.198 %,0.00 %,1.897 %,1.905 %,2.561 % .2. Influence on dissolvability of catalpol in Huanglian Bolus of different compatibility ratio of Rhizoma Coptidis and dried Rehmannia root A effective,stable,reliable method to detect the content of catalpol was established . The repetition ratio were 0 %,1.18 %,4.20 %,3.85 %,2.30 % and the dissolvability ratio of DR : RC 1: 0,0:1,1:1,1:4,1:8 were respectively 0.00 %,0.85 %,0.41 %,0.45 %,0.50 % .Conclusion1. Different compatibility Ratios of Rhizoma Coptidis ( RC ) and dried Rehmannia root ( DR ) could contribute to improve glucose and lipids profiles of type 1 diabetes models and reduced pancreatic pathological damage. In total , RC: DR 1:8 was more favorable and superior than other groups .2. RC with medium dosage of DR ( 1:1, 1:4 ) failed to increase the dissolvability of berberine while the increased dissolvability of berberine could be achieved due to massive dosage of RC with DR ( 1:8 ) . RC with DR could contribute to the decreased dissolvability of catalpol .
