Objectives To detect the interaction of p55γPI3K-Rb in cell line MKN-28 and tissues of gastrointestinal normal mucosa, paraneoplastic and gastrointestinal cancers and to elucidate its significance.Methods Proteins of MKN-28 cells were extracted. 20 cases of gastrointestinal normal mucosa, paraneoplastic and gastrointestinal cancers were carried out by rubbing. Immunoprecipitation was performed with anti-Rb antibody and Western Blot was performed with anti-p55γPI3K antibody to detect the interaction of p55γPI3K-Rb.Results (1) P55γPI3K-Rb pathway is expressed in gastric cancer cell line MKN-28. (2) In gastrointestinal normal mucosa, paraneoplastic and gastrointestinal cancers, expression of pRb is positive. Moreove, the expression of pRb in gastrointestinal cancers is higher than others(p<0.05). (3)In 20 cases of gastrointestinal tumor specimens 8 cases of p55γPI3K-Rb interaction are positive. And the interaction has no difference in tissues of gastrointestinal normal mucosa, paraneoplastic and gastrointestinal cancers (p>0.05).Conclusion Interaction of p55γPI3K-Rb exists in gastrointestinal tumors. It is possible to interrupt tumors genesis by blocking the interaction of p55γPI3K-Rb, which may provide a new treatment pathway for gastrointestinal cancers.
