| objective: The objective is to observe the expression of HOXC4 gene,HOXC6 gene on the differentiation and proliferation of hematopoietic stem cell to colony Colony Forming Unit-T Lymphocyte in vitro. And the differentiation progress affected by HCMV and/or ATRA, and to explore the possible mechanism of HCMV-induced maldevelopment to human cord blood T-Lymphocyte progenitor in genic level. Methods Normal CFU–TL culture was used as blank control , After MTT detection , the human cord blood Lymphocyte progenitor HOXC4,HOXC6 gene mRNA expression level after HCMV infection and/or ATRA were detected by Fluorogenic quantitative reserve transcription polymerize chain reaction(FQ-RT-PCR) method. Results HCMV of 106 PFU/ml was dilutedto 0.1ml 105 PFU /ml and added into the infected group. The expression of HOXC4 and HOXC6 genes in the differentiation process increased slightly on 3rd day ,and were up to the most on 7th day, (P〈0.05), but became lower in 12th day respectively in normal group, HCMVgroup,ATRA group,HCMV and ATRA group. Compared with the expression of HOXC6,the expression of HOXC4 was obviously higher in each group. Compared with the expression of HOXC4 and HOXC6 genes of normal group, the expression of HOXC4 and HOXC6 of the group ATRA were up-regulated remarkably (P<0.05), the expression of HOXC4 and HOXC6 of the group HCMV were down-regulated (P<0.05). Conclusion:1The expression of HOXC4 and HOXC6 genes mRNA appeared on regularly changes .There has a positive relationship between HOXC4,HOXC6 genes and lymphocyte progenitor hematopoiesis. 2. Compared with the expression of HOXC6 gene, the expression of HOXC4 gene was obviously higher in each group.HOXC4 gene was regarded as a main gene in the manipulation of in the proliferation and differentiation of lymphocyte progenitor cell. 3.HCMV can down-regulate the expression of HOXC4 and HOXC6 genes and lead a suppression effect on the cell morphology, which confirm the theory that the normal hematopoietic lineage determination and maturation rely on the stable and consistently expression of Homeobox genes. At the same condition , 6×10-8mol/l ATRA can up-regulate the expression of HOXC4 and HOXC6 genes. |
